Product Overview
This rabbit polyclonal antibody detects sphingosine kinase 1 (gene SPHK1) in human samples and is validated for Western blot, flow cytometry and ELISA. The predicted molecular weight is 42.5 kDa, and the supplier reports an observed band at about 52 kDa.
Sphingosine kinase 1 (SPHK1) phosphorylates sphingosine to produce sphingosine-1-phosphate, a lipid mediator that acts both inside cells and through S1P receptors on the cell surface. By lowering ceramide and raising S1P, SPHK1 generally favors cell growth and survival, and it is studied in inflammation, vascular biology and cancer.

Samples with a confirmed band (WB): human HeLa cells, human HepG2 cells, human MCF-7 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with a confirmed signal (flow cytometry): HepG2 cells.

Recombinant human SPHK1 fragment (Glu55–Asn357), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human. UniProt places sphingosine kinase 1 in the cytoplasm, nucleus, cell membrane and endosome membrane. Tissue expression noted by UniProt (human): Widely expressed with highest levels in adult liver, kidney, heart and skeletal muscle. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
