Biological background

Protein function (datasheet): Destroys radicals which are normally produced within the cells and which are toxic to biological systems.

Scientific background (datasheet): Superoxide dismutases (SOD) are a class of enzymes that catalyze the dismutation of superoxide into oxygen and hydrogen peroxide. As such, they are an important antioxidant defense in nearly all cells exposed to oxygen. One of the exceedingly rare exceptions is Lactobacillus plantarum and related lactobacilli, which use a different mechanism.Cu,Zn-SOD was found widely distributed in the cell cytosol and in the cell nucleus, consistent with it being a soluble cytosolic protein. Mitochondria and secretory compartments did not label for this protein. In human cells, peroxisomes showed a labeling density slightly less than that of cytoplasm.

Cellular localization (datasheet): Cytoplasm. Nucleus. Predominantly cytoplasmic; the pathogenic variants ALS1 Arg-86 and Ala-94 gradually aggregates and accumulates in mitochondria.

Tissue details (datasheet): Expressed in neurons of the rolandic area of the brain (at protein level). Expressed in the brain.

Sequence similarities (datasheet): Belongs to the Cu-Zn superoxide dismutase family.

Research relevance and current trends

  • Commonly studied in contexts related to Cancer,Cancer Metabolism,Cell Biology,Chaperones,Heat Shock Proteins,Metabolism,Oxidative Stress,Pathways and Processes,Protein Trafficking,Redox Metabolism,Signal Transduction.
  • Supports comparative expression analysis across conditions, genotypes, or treatments when paired with appropriate controls.
  • Useful for confirming target presence and subcellular distribution using orthogonal readouts (e.g., microscopy vs. immunoblotting).

Common research applications

  • Western blot (WB): Compare relative target abundance and apparent size/isoforms across samples; interpret bands in light of expected MW and potential PTMs.
  • ELISA: Measure target abundance in compatible matrices using a standard-curve readout; ensure dilution linearity and appropriate controls.
  • Immunohistochemistry (IHC): Assess tissue distribution and cell-type patterns; interpret staining with appropriate negative controls and antigen context.
  • Immunofluorescence / ICC: Visualize subcellular localization and co-localization patterns; consider fixation/permeabilization compatibility and controls.
  • Flow cytometry: Quantify target-positive populations in single-cell suspensions; pair with viability and isotype/FMO controls conceptually.

Notes for experimental interpretation

  • Consider isoforms, post-translational modifications, and processing that can shift apparent molecular weight or localization.
  • Cross-reactivity (datasheet): No cross-reactivity with other proteins
  • Use appropriate positive and negative controls (e.g., KO/KD, blocking peptide, or isotype controls) to support specificity interpretation.

As a polyclonal antibody, this reagent may recognize multiple epitopes on the target, which can improve detection robustness but may require careful specificity controls.

Does anti-Superoxide Dismutase 1/SOD1 antibody PA1345 work on pig ICC with fetal brain cortex?
Our lab technicians have not tested anti-Superoxide Dismutase 1/SOD1 antibody PA1345 on pig. You can run a BLAST between pig and the immunogen sequence of anti-Superoxide Dismutase 1/SOD1 antibody PA1345 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated pig samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in pig fetal brain cortex in ICC, you can get your next antibody for free.
Is there a BSA free version of anti-Superoxide Dismutase 1/SOD1 antibody PA1345 available?
Thanks for your recent telephone inquiry. I can confirm that some lots of this anti-Superoxide Dismutase 1/SOD1 antibody PA1345 are BSA free. For now, these lots are available and we can make a BSA free formula for you free of charge. It will take 3 extra days to prepare. If you require this antibody BSA free again in future, please do not hesitate to contact me and I will be pleased to check which lots we have in stock that are BSA free.
