Key elements and design rationale
- Antibody format: Rabbit Polyclonal Rabbit IgG
- Immunogen / epitope context: A synthetic peptide corresponding to a sequence at the C-terminus of human Synaptotagmin 1, identical to the related mouse and rat sequences.
- Molecular weight context: reported MW: 65 kDa; calculated MW: 33927 MW
- Reactivity: Human,Mouse,Rat
- Applications: Flow Cytometry, IHC, ICC, WB
As a polyclonal antibody, the reagent recognizes multiple epitopes on the target, which can improve detection robustness but may increase sensitivity to sample-dependent epitope changes.
Biological background
Adrenocorticotropic hormone receptor; synaptotagmin 1. Synaptotagmin-1 is a protein that in humans is encoded by the SYT1 gene. The synaptotagmins are integral membrane proteins of synaptic vesicles thought to serve as Ca (2+) sensors in the process of vesicular trafficking and exocytosis. Calcium binding to synaptotagmin-1 participates in triggering neurotransmitter release at the synapse. Functional note: May have a regulatory role in the membrane interactions during trafficking of synaptic vesicles at the active zone of the synapse. It binds acidic phospholipids with a specificity that requires the presence of both an acidic head group and a diacyl backbone. A Ca (2+)-dependent interaction between synaptotagmin and putative receptors for activated protein kinase C has also been reported. It can bind to at least three additional proteins in a Ca (2+)-independent manner; these are neurexins, syntaxin and AP2. Plays a role in dendrite formation by melanocytes (PubMed:23999003). Reported localization: Cytoplasmic vesicle, secretory vesicle membrane. Expression/tissue context: Expressed in melanocytes (PubMed:23999003).
Research relevance and current trends
- Neuroscience: Researchers commonly examine how SYT1 (Adrenocorticotropic hormone receptor) relates to this theme using model systems and orthogonal readouts.
- Neurotransmission: Researchers commonly examine how SYT1 (Adrenocorticotropic hormone receptor) relates to this theme using model systems and orthogonal readouts.
- Protein Trafficking: Researchers commonly examine how SYT1 (Adrenocorticotropic hormone receptor) relates to this theme using model systems and orthogonal readouts.
Common research applications
- Western blotting: compare relative SYT1 (Adrenocorticotropic hormone receptor) levels across conditions; band patterns may reflect isoforms and processing.
- IHC/IHC-F: assess spatial distribution of SYT1 (Adrenocorticotropic hormone receptor) across tissue regions and cell types using matched controls.
- IF/ICC: evaluate subcellular localization and co-localization patterns; signal can depend on fixation/permeabilization and epitope accessibility.
- Flow cytometry: quantify target-positive populations and shifts in expression; gating strategy and background staining controls are essential.
Notes for experimental interpretation
- Specificity notes: No cross reactivity with other proteins.
- Cross-reactivity: No cross-reactivity with other proteins.
- Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.
- Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.
