Biological background
Protein function (datasheet): Mediates homophilic cell-cell adhesion in a Ca (2+)- independent manner. Also mediates heterophilic cell-cell adhesion with CADM3 and PVRL3 in a Ca (2+)-independent manner. Acts as a tumor suppressor in non-small-cell lung cancer (NSCLC) cells. Interaction with CRTAM promotes natural killer (NK) cell cytotoxicity and interferon-gamma (IFN-gamma) secretion by CD8+ cells in vitro as well as NK cell-mediated rejection of tumors expressing CADM3 in vivo. May contribute to the less invasive phenotypes of lepidic growth tumor cells. In mast cells, may mediate attachment to and promote communication with nerves. CADM1, together with MITF, is essential for development and survival of mast cells in vivo. Acts as a synaptic cell adhesion molecule and plays a role in the formation of dendritic spines and in synapse assembly (By similarity). May be involved in neuronal migration, axon growth, pathfinding, and fasciculation on the axons of differentiating neurons. May play diverse roles in the spermatogenesis including in the adhesion of spermatocytes and spermatids to Sertoli cells and for their normal differentiation into mature spermatozoa. .
Scientific background (datasheet): Cell adhesion molecule 1 is a protein that, in humans, is encoded by the CADM1 gene. This gene is also known as IGSF4, SYNCAM or TSLC1, and it is mapped to 11q23.3. CADM1 functions as a homophilic cell adhesion molecule at the synapse. Expression of the isolated cytoplasmic tail of CADM1 in neurons inhibited synapse assembly. Conversely, expression of full-length CADM1 in nonneuronal cells induced synapse formation by cocultured hippocampal neurons with normal release properties. CADM1 has been shown to interact with EPB41L3.
Cellular localization (datasheet): Cell membrane ; Single-pass type I membrane protein . Cell junction, synapse . Associates with perinuclear and plasma membranes in vivo. Localized to the basolateral plasma membrane of epithelial cells in gall bladder. .
Tissue details (datasheet): Expressed in pancreatic islets and in brain, where it has been seen in cerebral cortex, hippocampus, basal ganglia, habenula and thalamus. Expressed in the small cell lung carcinoma cell line SCC-9. No expression in skeletal muscle. .
Sequence similarities (datasheet): Belongs to the nectin family.
Research relevance and current trends
- Commonly studied in contexts related to Integration Of Energy,Integration Of Energy Metabolism,Metabolic Signaling Pathway,Metabolic Signaling Pathways,Metabolism,Neuroscience,Neurotransmission,Pathways and Processes,Signal Transduction,Signaling Pathway.
- Supports comparative expression analysis across conditions, genotypes, or treatments when paired with appropriate controls.
- Useful for confirming target presence and subcellular distribution using orthogonal readouts (e.g., microscopy vs. immunoblotting).
Common research applications
- Western blot (WB): Compare relative target abundance and apparent size/isoforms across samples; interpret bands in light of expected MW and potential PTMs.
- ELISA: Measure target abundance in compatible matrices using a standard-curve readout; ensure dilution linearity and appropriate controls.
- Immunohistochemistry (IHC): Assess tissue distribution and cell-type patterns; interpret staining with appropriate negative controls and antigen context.
- Flow cytometry: Quantify target-positive populations in single-cell suspensions; pair with viability and isotype/FMO controls conceptually.
Notes for experimental interpretation
- Consider isoforms, post-translational modifications, and processing that can shift apparent molecular weight or localization.
- Cross-reactivity (datasheet): No cross-reactivity with other proteins
- Use appropriate positive and negative controls (e.g., KO/KD, blocking peptide, or isotype controls) to support specificity interpretation.
As a polyclonal antibody, this reagent may recognize multiple epitopes on the target, which can improve detection robustness but may require careful specificity controls.
