Overview

This antibody is intended for detection of Tbp (Tumor necrosis factor receptor superfamily member 1A) in biological samples using common immunoassay formats. It is typically selected based on target identity, species reactivity, clonality/clone information, and detection modality.

Vendor notes: Boster Bio Anti-TATA binding protein TBP Antibody Picoband® catalog # A00302. Tested in ELISA, WB applications. This antibody reacts with Mouse, Rat. The brand Picoband indicates this is a premium antibody that guarantees superior quality, high affinity, and strong signals with minimal background in Western blot applications. Only our best-performing antibodies are designated as Picoband, ensuring unmatched performance.

Key elements and design rationale

  • Antibody format: Rabbit Polyclonal Rabbit IgG
  • Immunogen / epitope context: E. coli-derived mouse Tbp recombinant protein (Position: L38-A107). (reported region: L38-A107).
  • Molecular weight context: reported MW: 38 kDa; calculated MW: 50495 MW
  • Reactivity: Mouse,Rat
  • Applications: ELISA, WB

As a polyclonal antibody, the reagent recognizes multiple epitopes on the target, which can improve detection robustness but may increase sensitivity to sample-dependent epitope changes.

Biological background

Tumor necrosis factor receptor superfamily member 1A; TATA box binding protein. TBP, TATA-binding protein, is a general transcription factor that binds specifically to a DNA sequence called the TATA box. The TBP gene is mapped to 6q27. This DNA sequence is found about 35 base pairs upstream of the transcription start site in some eukaryotic gene promoters. TBP, along with a variety of TBP-associated factors, make up the TFIID, a general transcription factor that in turn makes up part of the RNA polymerase II preinitiation complex. As one of the few proteins in the preinitiation complex that binds DNA in a sequence-specific manner, it helps position RNA polymerase II over the transcription start site of the gene. However, it is estimated that only 10-20% of human promoters have TATA boxes. Therefore, TBP is probably not the only protein involved in positioning RNA polymerase II. Functional note: General transcription factor that functions at the core of the DNA-binding multiprotein factor TFIID. Binding of TFIID to the TATA box is the initial transcriptional step of the pre- initiation complex (PIC), playing a role in the activation of eukaryotic genes transcribed by RNA polymerase II. Component of a BRF2-containing transcription factor complex that regulates transcription mediated by RNA polymerase III. Component of the transcription factor SL1/TIF-IB complex, which is involved in the assembly of the PIC (pre-initiation complex) during RNA polymerase I-dependent transcription. The rate of PIC formation probably is primarily dependent on the rate of association of SL1 with the rDNA promoter. SL1 is involved in stabilization of nucleolar transcription factor 1/UBTF on rDNA. Reported localization: Nucleus. Expression/tissue context: Ubiquitously expressed.

Research relevance and current trends

  • 2339: Researchers commonly examine how Tbp (Tumor necrosis factor receptor superfamily member 1A) relates to this theme using model systems and orthogonal readouts.
  • Epigenetics and Nuclear Signaling: Researchers commonly examine how Tbp (Tumor necrosis factor receptor superfamily member 1A) relates to this theme using model systems and orthogonal readouts.
  • Pol II Transcription: Researchers commonly examine how Tbp (Tumor necrosis factor receptor superfamily member 1A) relates to this theme using model systems and orthogonal readouts.

Common research applications

  • Western blotting: compare relative Tbp (Tumor necrosis factor receptor superfamily member 1A) levels across conditions; band patterns may reflect isoforms and processing.
  • ELISA-compatible use: when applicable, interpret signal as relative abundance across sample sets with consistent handling and dilution strategy.

Notes for experimental interpretation

  • Specificity notes: No cross reactivity with other proteins.
  • Cross-reactivity: No cross-reactivity with other proteins.
  • Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.
  • Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.

We have seen staining in mouse uterus. Are there any suggestions? Is anti-TATA binding protein TBP antibody supposed to stain uterus positively?
Based on literature uterus does express TBP. Based on Uniprot.org, TBP is expressed in forebrain, fibroblast, uterus, among other tissues. Regarding which tissues have TBP expression, here are a few articles citing expression in various tissues: Fibroblast, Pubmed ID: 2194289 Uterus, Pubmed ID: 14702039
I am interested in to test anti-TATA binding protein TBP antibody A00302 on mouse forebrain for research purposes, then I may be interested in using anti-TATA binding protein TBP antibody A00302 for diagnostic purposes as well. Is the antibody suitable for diagnostic purposes?
The products we sell, including anti-TATA binding protein TBP antibody A00302, are only intended for research use. They would not be suitable for use in diagnostic work. If you have the means to develop a product into diagnostic use, and are interested in collaborating with us and develop our product into an IVD product, please contact us for more discussions.
