Overview

This antibody is intended for detection of TNF (Tumor necrosis factor) in biological samples using common immunoassay formats. It is typically selected based on target identity, species reactivity, clonality/clone information, and detection modality.

Vendor notes: Boster Bio Anti-TNF alpha Antibody Picoband® catalog # PB9246. Tested in WB applications. This antibody reacts with Mouse. The brand Picoband indicates this is a premium antibody that guarantees superior quality, high affinity, and strong signals with minimal background in Western blot applications. Only our best-performing antibodies are designated as Picoband, ensuring unmatched performance.

Key elements and design rationale

  • Antibody format: Rabbit Polyclonal Rabbit IgG
  • Immunogen / epitope context: E.coli-derived mouse TNF alpha recombinant protein (Position: L80-Q227). Mouse TNF alpha shares 79% and 95% amino acid (aa) sequence identity with human and rat TNF alpha, respectively. (reported region: L80-Q227).
  • Molecular weight context: reported MW: 25 kDa; calculated MW: 25896 MW
  • Reactivity: Mouse
  • Applications: WB

As a polyclonal antibody, the reagent recognizes multiple epitopes on the target, which can improve detection robustness but may increase sensitivity to sample-dependent epitope changes.

Biological background

Tumor necrosis factor; Tumor necrosis factor. TNFα (Tumor Necrosis Factor alpha) gene encodes a multifunctional proinflammatory cytokine that belongs to the tumor necrosis factor (TNF) superfamily. This cytokine is mainly secreted by macrophages. It can bind to, and thus functions through its receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. This cytokine is involved in the regulation of a wide spectrum of biological processes including cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. This cytokine has been implicated in a variety of diseases, including autoimmune diseases, insulin resistance, and cancer. Knockout studies in mice also suggested the neuroprotective function of this cytokine. Functional note: Cytokine that binds to TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. It is mainly secreted by macrophages and can induce cell death of certain tumor cell lines. It is potent pyrogen causing fever by action or by stimulation of interleukin-1 secretion and is implicated in the induction of cachexia, Under certain conditions it can stimulate cell proliferation and induce cell differentiation. Reported localization: Cell membrane; Single-pass type II membrane protein. Expression/tissue context: Most abundant in placenta and skeletal muscle. It is also detected in the heart, lung, liver and kidney. IL15- S21AA is preferentially expressed in tissues such as testis and thymus.

Research relevance and current trends

  • Atherosclerosis: Researchers commonly examine how TNF (Tumor necrosis factor) relates to this theme using model systems and orthogonal readouts.
  • Cancer: Researchers commonly examine how TNF (Tumor necrosis factor) relates to this theme using model systems and orthogonal readouts.
  • Cardiovascular: Researchers commonly examine how TNF (Tumor necrosis factor) relates to this theme using model systems and orthogonal readouts.

Common research applications

  • Western blotting: compare relative TNF (Tumor necrosis factor) levels across conditions; band patterns may reflect isoforms and processing.

Notes for experimental interpretation

  • Specificity notes: No cross reactivity with other proteins.
  • Cross-reactivity: No cross-reactivity with other proteins
  • Family / similarity context: Belongs to the IL-15/IL-21 family.
  • Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.
  • Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.

Our team were satisfied with the WB result of your anti-TNF alpha antibody. However we have observed positive staining in prostatic carcinoma cell membrane using this antibody. Is that expected? Could you tell me where is TNF supposed to be expressed?
From what I have seen in literature, prostatic carcinoma does express TNF. Generally TNF expresses in cell membrane. Regarding which tissues have TNF expression, here are a few articles citing expression in various tissues: Blood, Pubmed ID: 15489334 Prostatic carcinoma, Pubmed ID: 8597870, 10205166
