Product Overview
This rabbit polyclonal antibody recognizes E3 ubiquitin-protein ligase TRAF7 in human samples and is validated for Western blot, flow cytometry and ELISA. The predicted molecular weight is 74.6 kDa, and the supplier reports an observed band at about 75 kDa.
TRAF7 is an atypical member of the TRAF family that combines a RING domain with WD40 repeats instead of the usual TRAF domain. It acts as an E3 ligase for ubiquitin and SUMO and influences MAP3K3-driven JNK/AP-1 signaling, innate immunity and apoptosis. TRAF7 mutations are frequent in meningiomas and also cause a developmental disorder.

Validated Applications
| Western blot | 0.1–0.25 µg/mL (Human) |
|---|---|
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human MCF-7 cells, human HeLa cells, human Caco-2 cells, human T-47D cells, human A549 cells, human Jurkat cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.25 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with a confirmed signal (flow cytometry): U87 cells.
Immunogen
Recombinant human TRAF7 fragment (Arg29–Cys670), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human. UniProt places E3 ubiquitin-protein ligase TRAF7 in the cytoplasmic vesicle, cytoplasm and nucleus. Tissue expression noted by UniProt (human): Ubiquitously expressed with high levels in skeletal muscle, heart, colon, spleen, kidney, liver and placenta. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
