Vendor description (summary): Boster Bio Anti-TRPV3 Antibody Picoband® catalog # A03874-1.
Biological background
Biological context: Putative receptor-activated non-selective calcium permeant cation channel. It is activated by innocuous (warm) temperatures and shows an increased response at noxious temperatures greater than 39 degrees Celsius. Activation exhibits an outward rectification. May associate with TRPV1 and may modulate its activity. Is a negative regulator of hair growth and cycling: TRPV3-coupled signaling suppresses keratinocyte proliferation in hair follicles and induces apoptosis and premature hair follicle regression.
Expression and localization notes: cellular localization: Membrane. Multi-pass membrane protein., tissue context: Abundantly expressed in CNS. Widely expressed at low levels. Detected in dorsal root ganglion. Expressed in the keratinocyte layers of the outer root sheath and, to lesser extent, to the matrix of the hair follicles..
Common research applications
- Western blotting (WB): Compare TRPV3 levels across samples and conditions using appropriate loading and biological controls.
- Immunohistochemistry (IHC): Evaluate spatial distribution of TRPV3 in tissue sections, considering fixation and antigen retrieval effects.
- Immunofluorescence / ICC: Assess subcellular localization patterns and co-localization with compartment markers in cultured cells.
- Flow cytometry: Quantify TRPV3-positive populations in single-cell suspensions with appropriate gating and controls.
- ELISA: Use antibody-based detection formats to assess antigen presence or binding in plate-based assays.
Notes for experimental interpretation
- Account for isoforms, post-translational modifications, and sample-specific processing that can shift apparent molecular weight or epitope accessibility.
- Use positive/negative biological controls where possible (e.g., known-expressing cells/tissues, knockdown/knockout models) and include appropriate secondary-only/isotype controls for imaging workflows.
Additional product notes (from provided fields)
- Specificity: No cross reactivity with other proteins.
- Background: TRPV3 (Transient Receptor Potential Cation Channel Subfamily V Member 3), also known as VRL3, is a human gene encoding the protein of the same name. The TRPV3 protein belongs to a family of nonselective cation channels that function in a variety of processes, including temperature sensation and vasoregulation. Peier et al. localized the TRPV3 gene to a BAC clone mapped to chromosome 17p13. They mapped the mouse gene to chromosome 11B4. Peier et al. stably expressed mouse Trpv3 in Chinese hamster ovary cells and assayed electrophysiologic activity by whole cell voltage-clamp techniques. They determined that Trpv3 is a cation-permeable channel activated by warm and hot temperatures. Xu et al. showed that increasing temperature from approximately 22 to 40 degrees Celsius in mammalian cells transfected with human TRPV3 elevated intracellular calcium by activating a nonselective cationic conductance.
- Cross reactivity: No cross-reactivity with other proteins.
- Cellular localization: Membrane. Multi-pass membrane protein.
- Tissue details: Abundantly expressed in CNS. Widely expressed at low levels. Detected in dorsal root ganglion. Expressed in the keratinocyte layers of the outer root sheath and, to lesser extent, to the matrix of the hair follicles.
