Overview

This antibody is intended for detection of UBB (Polyubiquitin-B) in biological samples using common immunoassay formats. It is typically selected based on target identity, species reactivity, clonality/clone information, and detection modality.

Vendor notes: Boster Bio Anti-Ubiquitin/UBB Antibody Picoband® catalog # PB9122. Tested in IF, IHC, ICC, WB applications. This antibody reacts with Human, Mouse, Rat. The brand Picoband indicates this is a premium antibody that guarantees superior quality, high affinity, and strong signals with minimal background in Western blot applications. Only our best-performing antibodies are designated as Picoband, ensuring unmatched performance.

Key elements and design rationale

  • Antibody format: Rabbit Polyclonal Rabbit IgG
  • Immunogen / epitope context: E.coli-derived human Ubiquitin recombinant protein (Position: M77-G152). (reported region: M77-G152).
  • Molecular weight context: reported MW: 9 kDa; calculated MW: 25762 MW
  • Reactivity: Human,Mouse,Rat
  • Applications: IF, IHC, ICC, WB

As a polyclonal antibody, the reagent recognizes multiple epitopes on the target, which can improve detection robustness but may increase sensitivity to sample-dependent epitope changes.

Biological background

Polyubiquitin-B; Polyubiquitin-B. Ubiquitin (originally, ubiquitous immunopoietic polypeptide) is a small regulatory protein that has been found in almost all tissues (ubiquitously) of eukaryotic organisms. Its gene is mapped to 16p 13.3. The ubiquitin protein itself consists of 76 amino acids and has a molecular mass of about 8.5 kDa. Ubiquitin binds to proteins and labels them for destruction. It s protein recycling and also proteins to other locations in the cell, where they control other protein and cell mechanisms. Functional note: Ubiquitin: Exists either covalently attached to another protein, or free (unanchored). When covalently bound, it is conjugated to target proteins via an isopeptide bond either as a monomer (monoubiquitin), a polymer linked via different Lys residues of the ubiquitin (polyubiquitin chains) or a linear polymer linked via the initiator Met of the ubiquitin (linear polyubiquitin chains). Polyubiquitin chains, when attached to a target protein, have different functions depending on the Lys residue of the ubiquitin that is linked: Lys-6-linked may be involved in DNA repair; Lys-11-linked is involved in ERAD (endoplasmic reticulum-associated degradation) and in cell-cycle regulation; Lys-29-linked is involved in lysosomal degradation; Lys-33-linked is involved in kinase modification; Lys-48-linked is involved in protein degradation via the proteasome; Lys-63-linked is involved in endocytosis, DNA-damage responses as well as in signaling processes leading to activation of the transcription factor NF-kappa-B. Linear polymer chains formed via attachment by the initiator Met lead to cell signaling. Ubiquitin is usually conjugated to Lys residues of target proteins, however, in rare cases, conjugation to Cys or Ser residues has been observed. When polyubiquitin is free (unanchored-polyubiquitin), it also has distinct roles, such as in activation of protein kinases, and in signaling. . Reported localization: Ubiquitin: Cytoplasm . Nucleus . Expression/tissue context: Liver, brain, heart, lymphocytes, fibroblasts, biliary epithelium, pancreas, skeletal muscle, kidney, lung and placenta.

Research relevance and current trends

  • Alzheimer's Disease: Researchers commonly examine how UBB (Polyubiquitin-B) relates to this theme using model systems and orthogonal readouts.
  • Chromatin Modifying Enzymes: Researchers commonly examine how UBB (Polyubiquitin-B) relates to this theme using model systems and orthogonal readouts.
  • Epigenetics and Nuclear Signaling: Researchers commonly examine how UBB (Polyubiquitin-B) relates to this theme using model systems and orthogonal readouts.

Common research applications

  • Western blotting: compare relative UBB (Polyubiquitin-B) levels across conditions; band patterns may reflect isoforms and processing.
  • IHC/IHC-F: assess spatial distribution of UBB (Polyubiquitin-B) across tissue regions and cell types using matched controls.
  • IF/ICC: evaluate subcellular localization and co-localization patterns; signal can depend on fixation/permeabilization and epitope accessibility.

Notes for experimental interpretation

  • Specificity notes: No cross reactivity with other proteins.
  • Cross-reactivity: No cross-reactivity with other proteins
  • Family / similarity context: Belongs to the ubiquitin family.
  • Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.
  • Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.
