Product Overview
This rabbit polyclonal antibody recognizes ubiquitin carboxyl-terminal hydrolase 10 (gene USP10) in human, mouse and rat samples and is validated for Western blot and ELISA. The predicted molecular weight is 87.1 kDa; the supplier reports observed bands at 110 kDa, 130 kDa.
USP10 is a deubiquitinating enzyme best known for stabilizing the tumor suppressor p53 by opposing MDM2-driven ubiquitination, including after DNA damage. It also removes ubiquitin from ribosomal proteins, Beclin 1 and CFTR, linking it to ribosome quality control and autophagy. USP10 is studied in stress responses and cancer.

Validated Applications
| Western blot | 0.1–0.25 µg/mL (Human, Mouse, Rat) |
|---|---|
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human HeLa cells, human 293T cells, human MCF-7 cells, human A549 cells, human K562 cells, human SIHA cells, rat brain tissue, mouse brain tissue, mouse NIH/3T3 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.25 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Immunogen
Recombinant human USP10 fragment (Met1–Leu798), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places ubiquitin carboxyl-terminal hydrolase 10 in the cytoplasm, nucleus and early endosome. Tissue expression noted by UniProt (human): Widely expressed. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
