Product Overview
Clone 3G11G2 is a mouse monoclonal antibody that recognizes 14-3-3 protein epsilon (gene YWHAE) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF and flow cytometry. The predicted molecular weight is 29.2 kDa, and the supplier reports an observed band at about 29 kDa.
14-3-3 ε (YWHAE) is a member of the 14-3-3 family of phospho-binding adapter proteins that regulate many signaling pathways. By binding phosphorylated partners it influences their activity and localization in processes such as cell-cycle checkpoints, apoptosis and neuronal migration, and deletion of YWHAE contributes to the severity of Miller-Dieker lissencephaly.

Samples with a confirmed band (WB): human HeLa cells, human SH-SY5Y cells, human SW620 cells, human Raji cells, rat brain tissue, rat PC-12 cells, mouse brain tissue, mouse NIH/3T3 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with confirmed staining (IHC): human colorectal adenocarcinoma tissue, human liver cancer tissue, human tonsil tissue, human endometrial cancer tissue, human placenta tissue, human bladder epithelial carcinoma tissue, human laryngeal squamous cells carcinomas tissue, rat brain tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): Caco-2 cells.
Samples with a confirmed signal (flow cytometry): ANA-1 cells, NRK cells.

Recombinant human YWHAE fragment (Met1–Gln255), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places 14-3-3 protein epsilon in the nucleus, cytoplasm and melanosome. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
