Biological Activity

E197 is a DOCK5 inhibitor. E197 can inhibit both mouse and human osteoclast activity. In particular, E197 prevented pathological bone loss in mice. Most interestingly, treatment with E197 did not affect osteoclast and osteoblast numbers and hence did not impair bone formation. E197 could represent a lead molecule to develop new antiosteoporotic drugs targeting the mechanism of osteoclast adhesion onto the bone.

Can high-temperature sterilization be used for inhibitors?

Most small molecule inhibitors are not compatible with autoclaving (121 °C), which can cause thermal decomposition or loss of biological activity. For sterile cell-based experiments, filter-sterilize working solutions through a 0.2 µm membrane after dilution into aqueous buffer. Verify thermal stability in the DataSheet before applying any heat treatment.

Animals cannot tolerate DMSO well. How should the dosage of DMSO be controlled during administration?

For normal mice, the concentration of DMSO should be kept below 10%. For nude mice, transgenic mice, or mice with weak tolerance, the concentration of DMSO should be kept below 2%. For inhibitors being used for the first time, it is recommended to perform a solvent-negative control experiment to confirm that the solvent has no nonspecific effects on the animals.
