Scientific Background
Zaire Ebola virus (EBOV) is the most lethal species of the Ebolavirus genus, responsible for multiple large outbreaks including the 2014–2016 West Africa epidemic. EBOV is classified as a BSL-4 pathogen, with case fatality rates of up to 90% in untreated patients. The viral glycoprotein (GP) mediates cell entry via macropinocytosis and NPC1-dependent membrane fusion. Safe surrogate systems for studying EBOV entry and evaluating anti-EBOV countermeasures are critical for biodefense and therapeutic development.

This HA-EBOV VLP is a hybrid alpha-pseudovirus assembled from Ebola Virus Zaire structural proteins (GP, VP40, NP) with an alphavirus-derived reporter RNA payload. BSL-2 safe and ready to use for studying viral entry. Available with Firefly luciferase, GFP, or RFP reporter.

Safety & Handling
Yes. HA-EBOV VLPs are single-cycle pseudoviruses that cannot produce infectious Ebola virus. They can be handled under standard BSL-2 containment, enabling entry and neutralization studies without BSL-4 requirements.
Vero or other permissive cells are infected with HA-EBOV VLPs or a VLP/test-sample mixture for ~1 hr. After overnight incubation, reporter expression (luciferase or fluorescent protein) is quantified. Neutralization is measured as percentage inhibition relative to virus-only controls.
VeroE6 cells are commonly used for filovirus pseudovirus assays. HEK293T cells expressing the NPC1 receptor can also be used for enhanced transduction efficiency in some protocols.
