Product Overview

This synthetic EHD1 Blocking Peptide corresponds to the immunogen sequence used to generate the paired anti-EHD1 antibody (STJ500849). Pre-absorbing the antibody with this peptide before use demonstrates epitope-specific binding and confirms that observed signals in Western blot, IHC, or IF are attributable to the target protein, not antibody cross-reactivity.

Pre-Absorption Specificity Protocol

  1. Dilute the anti-EHD1 antibody to the recommended working concentration in assay buffer.
  2. Add the blocking peptide at a 5–10× molar excess relative to the antibody. For typical IHC conditions (2 μg/mL antibody): use 10–20 μg/mL blocking peptide.
  3. Incubate at room temperature for 1 hour (or at 4 °C overnight) with gentle mixing.
  4. Use the pre-absorbed mixture as the antibody solution in your standard WB/IHC/IF protocol, run in parallel with a non-blocked antibody control.
  5. Complete loss of signal in the pre-absorbed lane confirms epitope-specific binding. A non-specific irrelevant peptide should be included as a negative control.

How do I use this EHD1 blocking peptide to confirm antibody specificity?

Pre-incubate the anti-EHD1 antibody with this peptide at 5–10× molar excess (e.g., 5–10 μg peptide per 1–2 μg antibody) for 1–2 h at room temperature. Run parallel blots — one with antibody alone (positive), one with antibody + peptide. Loss of signal in the peptide-blocked lane confirms specificity.

What peptide concentration should I use for blocking?

Start at 5–10 μg/mL peptide alongside 1–2 μg/mL antibody. Increase to 20× molar excess if partial signal persists. For IHC, pre-incubation at 4 °C overnight improves blocking efficiency.

Which antibody does this EHD1 peptide pair with?

This peptide is designed to pair with antibody STJ503996. Immunogen region: Synthetic peptide taken wtihin amino acid region 515-565 on human EH-domain containing protein 1.. Using with a different antibody clone may not produce blocking results if epitopes differ.

What applications is this peptide validated for?

Validated for Immunodepletion specificity controls. Performance in other applications (ELISA, IP) requires independent validation.

How should I store this peptide and what is its shelf life?

Store at-20°C for long term storage. Avoid freeze-thaw cycles.. Prepare single-use aliquots to avoid freeze-thaw; stable ≥12 months at −20 °C when lyophilized.
