Pre-Absorption Specificity Protocol
- Dilute the anti-EHD1 antibody to the recommended working concentration in assay buffer.
- Add the blocking peptide at a 5–10× molar excess relative to the antibody. For typical IHC conditions (2 μg/mL antibody): use 10–20 μg/mL blocking peptide.
- Incubate at room temperature for 1 hour (or at 4 °C overnight) with gentle mixing.
- Use the pre-absorbed mixture as the antibody solution in your standard WB/IHC/IF protocol, run in parallel with a non-blocked antibody control.
- Complete loss of signal in the pre-absorbed lane confirms epitope-specific binding. A non-specific irrelevant peptide should be included as a negative control.

Pre-incubate the anti-EHD1 antibody with this peptide at 5–10× molar excess (e.g., 5–10 μg peptide per 1–2 μg antibody) for 1–2 h at room temperature. Run parallel blots — one with antibody alone (positive), one with antibody + peptide. Loss of signal in the peptide-blocked lane confirms specificity.
Start at 5–10 μg/mL peptide alongside 1–2 μg/mL antibody. Increase to 20× molar excess if partial signal persists. For IHC, pre-incubation at 4 °C overnight improves blocking efficiency.
