    Overview

For quantitatve determination of acetaldehyde enzyme activity and evaluation of drug modulators. The assay uses OD565nm for signal readout. Compatible sample input includes Biological samples (e.g. plasma, serum, urine, tissue, and culture media) and food/beverage samples (e.g. wine, coffee, and juice). Typical stated assay timing is 30 min.

    Key elements and design rationale

    • Readout format: OD565nm supports plate-based signal acquisition and consistent comparison across matched samples.
    • Sample compatibility: The stated sample scope includes Biological samples (e.g. plasma, serum, urine, tissue, and culture media) and food/beverage samples (e.g. wine, coffee, and juice), which is useful when aligning matrix type with calibration and control design.
    • Analytical range context: The supplied specifications include a stated detection limit of 2 µM for interpreting low-signal samples.
    • Feature emphasis: Fast and sensitive. Linear detection range (20µL sample): 2µM to 2 mM acetaldehyde in a 96-well plate assay.

Additional feature notes highlight High-throughput.The procedure involves adding a single working reagent and reading the absorbance after 30 minutes. Room temperature assay. No 37°C heater is needed; Convenient.Homogeneous “mix-incubate-measure” type assay. Can be readily automated on HTS liquid handling systems for processing thousands of samples per day. Available format information for this listing includes 100 Tests.

    Biological background

This product is centered on measurement of acetaldehyde within the matrices described for the assay. In practice, datasets from this type of format are typically interpreted by comparing relative signal, activity, or abundance across matched control and experimental groups rather than relying on a single value in isolation. Careful alignment of sample matrix, incubation window, and calibration strategy is important when comparing results across plates, operators, or study days.

    More details

ACETALDEHYDE (CH3CHO) is one of the most widely occurring aldehydes in nature and is commonly used in industry. Acetaldehyde, a metabolic byproduct of ethanol in the liver, is toxic to the human body and is rapidly converted to the less harmful acetic acid by the enzyme aldehyde dehydrogenase. People with a deficiency of aldehyde dehydrogenase accumulate acetaldehyde when consuming alcohol and this accumulation results in facial and body flushing often referred to as “Asian flush syndrome.” Build up of acetaldehyde has also been associated with the effects of hangovers from alcohol consumption. Although classified as a carcinogen, acetaldehyde is naturally found in many foods and beverages such as ripe fruit, coffee, and wine. BioAssay Systems colorimetric acetaldehyde assay kit is based on aldehyde dehydrogenase catalyzed oxidation of acetaldehyde, in which the formed NADH reduces a formazan reagent. The intensity of the product color, measured at 565 nm, is directly proportional to the acetaldehyde concentration in the sample.

    Detection method

    Colorimetric (OD 565 nm).

    Detection limit and analytical sensitivity

    Reported detection limit: 2 µM.

    Procedures and timing

Stated procedure or timing information: 30 min.

    Research relevance and current trends

    • Plate-based quantification and side-by-side group comparison remain central use cases for this assay format.
    • The product notes emphasize multi-sample throughput, making it relevant for screening-oriented and larger batch comparison studies.
    • The description supports intervention-focused study designs in which researchers compare baseline and perturbed conditions.

    Common research applications

    • Quantify acetaldehyde in plasma, serum, urine by OD565 nm readout.
    • Compare treatment or phenotype groups using matched plasma, serum, urine handling.
    • Monitor time-course or pre/post changes in plasma, serum, urine across study conditions.

Interpretation is usually strongest when signal changes are assessed alongside matrix-matched controls, replicate agreement, and the assay's stated analytical window.

    Notes for experimental interpretation

    • Matrix composition, background signal, and sample handling can influence apparent response; compare like-with-like whenever possible.
    • Use appropriate blanks, controls, and replicate wells to distinguish biological differences from plate, reagent, or handling variability.
How specific is this assay for acetaldehyde?

The assay mechanism uses the enzyme aldehyde dehydrogenase to quantify acetaldehyde in the sample. The enzyme will also oxidize other aldehydes in the sample (i.e. propionaldehyde). Therefore, the assay will not distinguish between acetaldehyde and other aldehydes present in the sample. Consideration should be taken if high levels of other aldehydes are expected in the sample being tested.

How long are the diluted standards good for?

Acetaldehyde is a rather volatile chemical, so proper storage of the standard is vital. Only bring the 3 M Standard to room temperature for time needed to prepare standards. Return to -20°C within 30 minutes of thawing. Be sure to keep the Standard and standard dilutions on ice or refrigerated with the tube caps closed during assay procedure. The diluted standards for the colorimetric assay (4 mM, 2.4 mM, 1.2 mM, and 0 mM) and the fluorimetric assay (60 µM, 36 µM, 18 µM, and 0 µM) should be prepared fresh before each assay and used within 10 minutes of preparing.

What temperature do I run the assay at?

The assay may be run at room temperature (25°C) or 37°C.
