Biological Activity
G-418 (Geneticin), an aminoglycoside antibiotic resembling gentamicin in structure, demonstrates toxicity to both eukaryotic and prokaryotic cells by obstructing protein synthesis (1).

When solubility is listed as < 1 mg/mL, the compound dissolves to less than 1 mg per milliliter in water at 25 °C, pH 7.4. Such compounds require an organic co-solvent such as DMSO to prepare a concentrated stock, which is then diluted into aqueous assay buffer. For cell-based assays, keep the final DMSO concentration below 0.1% (v/v) to avoid solvent-induced cytotoxicity.
Try the following steps: (1) warm the DMSO solvent to 37–50 °C before adding the compound; (2) vortex vigorously for 30 seconds then bath-sonicate for 1–2 minutes; (3) if still undissolved, prepare a more dilute stock.
