Target GABAA Receptor Alpha 5

The GABA-A receptor alpha 5 subunit (GABRA5) is a critical component of the GABA-A receptor complex, the brain’s primary fast-acting inhibitory neurotransmitter system. As part of the ligand-gated ion channel superfamily, GABRA5 is predominantly expressed in the hippocampus, where it plays a central role in modulating tonic inhibition, synaptic plasticity, and cognitive function.

Recent research has highlighted GABRA5 as a promising target in neurodegenerative disease, particularly Alzheimer’s disease. Inverse agonists selective for GABRA5-containing receptors have shown potential in enhancing learning and memory by reducing excessive inhibitory tone, which is often elevated in neurodegenerative states. This modulation of GABRA5 activity may help restore excitatory-inhibitory balance, improve cognitive performance, and slow disease progression.

Beyond Alzheimer’s, GABRA5 is also being investigated for its role in age-related cognitive decline and other neurological disorders characterized by impaired GABAergic signaling. Its unique pharmacological profile and region-specific expression make it an attractive candidate for precision therapies aimed at cognitive enhancement and neuroprotection.

As neuroscience advances toward targeted interventions, GABRA5 stands out as a key molecular player in the development of next-generation treatments for neurodegenerative diseases.

A 1:100 dilution of SMC-494 was sufficient for detection of GABA-A R, Alpha5 in 20 µg of mouse brain lysate by ECL immunoblot analysis using Goat anti-mouse IgG:HRP as the secondary antibody.
