The GABA-A receptor delta subunit (GABRD) is a specialized component of the GABA-A receptor family, which comprises fast-acting ligand-gated ion channels responsible for inhibitory neurotransmission in the brain. Unlike synaptic GABA-A receptors, delta-containing receptors are primarily located extrasynaptically and mediate tonic inhibition—providing a continuous inhibitory tone that stabilizes neural networks and prevents hyperexcitability.
GABRD is highly expressed in brain regions involved in cognition and emotional regulation, such as the thalamus, hippocampus, and cerebellum. Its unique role in maintaining baseline inhibitory control makes it a critical factor in neurological health. Dysregulation of GABRD has been implicated in a range of disorders, including epilepsy, anxiety, and sleep disturbances. More recently, its involvement in neurodegenerative diseases has gained attention, particularly in conditions like Alzheimer’s disease, where impaired tonic inhibition contributes to network dysfunction, excitotoxicity, and cognitive decline.
Targeting GABRD-containing receptors offers a promising therapeutic strategy for restoring inhibitory balance and protecting against neurodegeneration. As research advances, GABRD is emerging as both a biomarker and a molecular target in the development of precision treatments for GABAergic dysfunction in aging and disease.
2 µg/ml of SMC-345 was sufficient for detection of Delta1 GABA-A receptor in 10 µg of rat brain lysate by colorimetric immunoblot analysis using goat anti-mouse IgG:HRP as the secondary antibody.
