This kit uses a competitive inhibition ELISA format to quantitatively measure Gamma-Aminobutyric Acid (gABA) concentrations. In this assay, free analyte in the sample competes with immobilized antigen for a fixed amount of specific antibody. This kit uses a competitive inhibition ELISA format. Free Gamma-Aminobutyric Acid (gABA) in your sample competes with antigen immobilised on the microplate for binding to a specific detection antibody. The optical signal generated is inversely proportional to the concentration of analyte present — the higher the sample concentration, the lower the absorbance reading. A standard curve is used to convert OD values into absolute concentrations.
This kit has been validated for use with serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. Samples should be diluted appropriately within the assay detection range (31.2-2000pg/mL). Cell culture supernatants and tissue homogenates may require preliminary centrifugation to remove particulates. Avoid repeated freeze-thaw cycles — aliquot samples before freezing and use within the validated stability window. Haemolysed, lipaemic, or heavily precipitated samples may interfere with assay performance.
The detection range is 31.2-2000pg/mL with a minimum detectable concentration (sensitivity) of 10.9pg/mL. Samples with concentrations above the upper limit of the range should be diluted with the provided sample diluent and re-assayed. Samples falling below the lower limit may indicate undetectable levels or may require sample concentration prior to assay. Always run a standard curve in duplicate in every plate for accurate interpolation.
Traditional format contains concentrated reagents that require preparation prior to use (dilution of wash buffer, detection antibody, streptavidin-HRP, etc.). This format offers maximum flexibility and is preferred by experienced ELISA users who wish to customise incubation conditions or buffer composition. Ready-to-use format provides all working-strength reagents pre-prepared, significantly reducing hands-on time and minimising inter-operator variability. This format is ideal for routine clinical research, high-throughput laboratories, or users new to ELISA. Both formats use the same antibody pairs and deliver equivalent analytical performance.
Store all kit components at 2–8 °C upon receipt. Do not freeze reagents unless specified in the Instruction Manual, as this may compromise antibody activity and standard integrity. The microplate should be returned to its foil pouch with desiccant and resealed after each use. Shelf life: Traditional: 12 months | Ready-to-use: 16 months from manufacture date when stored under recommended conditions. Refer to the lot-specific Instruction Manual for the exact expiry date printed on the kit label.
