Assay Kit

Acetylation of histone H3 at lysine 9 (H3K9) is a post-translational modification where an acetyl group is added to the lysine residue at position 9 on the histone H3 protein. This modification is mediated by histone acetyltransferases (HATs), which add acetyl groups, and histone deacetylases (HDACs), which remove them. H3K9 acetylation is associated with gene activation. It is often found at active promoters and enhancers, facilitating a more open chromatin structure that allows transcription factors to access DNA. Altered acetylation levels at H3K9 have been linked to various cancers, affecting tumor progression and metastasis. The Global Acetyl Histone H3K9 Quantification Kit is designed to measure global histone H3K9 acetylation in various mammalian cells, including human and mouse cells, as well as fresh and frozen tissues, cultured adherent cells, and suspension cells. This assay is simple, user-friendly, accurate, and fast. The measurement can be completed in 2.5 hours, with a recommended sample input of 20 ng to 5 µg per well. Alternative Name: H3K9ac; Acetyl Histone H3K9; Acetylation of histone H3 at lysine 9

Assay Principle

The Global Acetyl Histone H3K9 Quantification Kit (Colorimetric) is a colorimetric immunoassay for the quantitative measurement of acetyl-histone h3k9 in mammalian cells and tissues. Target histones are captured on a pre-coated microplate and detected with a modification-specific antibody, then quantified against a standard curve. The assay measures global histone h3k9 acetylation with a recommended sample input of 20 ng – 5 µg per well and can be completed in 2.5 hours.

Kit Components

The kit provides a pre-coated microplate, target standard for standard-curve generation, detection reagents (modification-specific antibody and enzyme/fluorophore conjugate), wash buffer, and developing/stop solutions. Refer to the product datasheet for the full component list and volumes.
