Assay Principle

Sandwich ELISA

This kit uses a two-site sandwich ELISA to quantitate ANP (NPPA) in human samples. The plate is pre-coated with anti-ANP capture antibody. Samples and standards bind the capture antibody; biotinylated detection antibody and Streptavidin-HRP are applied; TMB substrate generates colorimetric signal at 450 nm proportional to ANP concentration. Sensitivity: 7.6 pg/mL; range: 15.6–1,000 pg/mL.

Scientific Background

Atrial Natriuretic Peptide (ANP), encoded by the NPPA gene (UniProt P01160), is a 28-amino acid cardiac hormone secreted by atrial cardiomyocytes in response to increased atrial wall stress (volume overload, elevated filling pressure). ANP exerts natriuretic, diuretic, and vasodilatory effects, opposing the renin-angiotensin-aldosterone system (RAAS) to reduce blood pressure and cardiac preload. Plasma ANP is markedly elevated in heart failure, atrial fibrillation, and hypertension, making it a validated cardiac biomarker alongside BNP and NT-proBNP. Also known as: ANF, ATFB6, CDD-ANF, PND; atrial natriuretic factor; atriopeptin; cardionatrin.

It promotes natriuresis, diuresis, and vasodilation to reduce cardiac preload. Plasma ANP is elevated in heart failure, atrial fibrillation, and hypertension, making it a clinically relevant research biomarker.

What is the detection sensitivity and what does it mean in practice?

Sensitivity is 7.6 pg/mL. Normal human plasma ANP is 20–77 pg/mL in healthy adults — comfortably within the detectable range (15.6–1,000 pg/mL) and well above the LLOD. Samples from heart failure or fluid overload subjects may exceed 100 pg/mL — prepare 2–5-fold dilutions if OD exceeds the upper standard.

What sample types are compatible with this assay?

Prepare an 8-point curve by 2-fold serial dilution of the provided ANP standard from 1,000 pg/mL to 15.6 pg/mL, plus a zero blank. Run in duplicate. Read at 450 nm (reference 570 nm or 540 nm). Fit with a 4-parameter logistic (4PL) model for accurate interpolation.

Can this kit be used alongside BNP or NT-proBNP assays?

Yes. ANP and BNP/NT-proBNP assays are independent and can be run on the same sample set in parallel. ANP reflects atrial stretch and preload; BNP/NT-proBNP primarily reflect ventricular wall stress. Running both provides a more complete picture of cardiac filling pressures and natriuretic peptide biology.
