Scientific Background
APLP1 encodes a member of the highly conserved amyloid precursor protein gene family. The encoded protein is a membrane-associated glycoprotein that is cleaved by secretases in a manner similar to amyloid beta A4 precursor protein cleavage. This cleavage liberates an intracellular cytoplasmic fragment that may act as a transcriptional activator. The encoded protein may also play a role in synaptic maturation during cortical development. Alternatively spliced transcript variants encoding different isoforms have been described.APLP1 and APLP2 double knockout mice display hypoglycemia and hyperin.
Assay Principle
This assay employs a two-site sandwich ELISA to quantitate B-APP in samples. An antibody specific for B-APP has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyB-APP present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for B-APP is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of B-APP bound in the initial step. The color development is stopped and the intensity of the color is measured.
