GPR109A is a high affinity receptor for nicotinic acid (niacin) and is a member of the nicotinic acid receptor family of G protein-coupled receptors (the other identified member being GPR109B).GPR109A is a Gi / Go protein-coupled receptor with high affinity for nicotinic acid.GPR109A is believed to be an important biomolecular target of niacin which is a widely prescribed drug for the treatment of dyslipidemia and to increase HDL cholesterol but whose therapeutic use is limited by flushing. Assay Principle
This assay employs a two-site sandwich ELISA to quantitate GPR109A in samples. An antibody specific for GPR109A has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyGPR109A present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for GPR109A is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of GPR109A bound in the initial step. The color development is stopped and the intensity of the color is measured.
