Assay Principle
HMGB1 (high mobility group box 1) is a highly conserved nuclear protein that acts as a chromatin-binding factor and, when released from damaged or activated cells, as a potent damage-associated molecular pattern (DAMP) driving inflammation. Circulating HMGB1 is studied as a biomarker in sepsis, sterile injury, autoimmunity, and cancer.
In this sandwich-format assay, a plate pre-coated with a capture antibody specific for HMGB1 (High mobility group protein B1) binds analyte from the sample; a biotinylated detection antibody and HRP-conjugate then form the detection complex, and color development is proportional to analyte concentration.

Assay Workflow
The kit provides the components required for standard-curve quantification, including pre-coated microplate strips, standards, conjugate, substrate, and wash and stop solutions (see the kit manual for the full component table). Absorbance is read on a standard microplate reader, and analyte concentrations are interpolated from the standard curve. Total hands-on-to-result time is approximately 3.5 hours.
