Assay Principle

HMGB1 (high mobility group box 1) is a highly conserved nuclear protein that acts as a chromatin-binding factor and, when released from damaged or activated cells, as a potent damage-associated molecular pattern (DAMP) driving inflammation. Circulating HMGB1 is studied as a biomarker in sepsis, sterile injury, autoimmunity, and cancer.

In this sandwich-format assay, a plate pre-coated with a capture antibody specific for HMGB1 (High mobility group protein B1) binds analyte from the sample; a biotinylated detection antibody and HRP-conjugate then form the detection complex, and color development is proportional to analyte concentration.

Assay Workflow

The kit provides the components required for standard-curve quantification, including pre-coated microplate strips, standards, conjugate, substrate, and wash and stop solutions (see the kit manual for the full component table). Absorbance is read on a standard microplate reader, and analyte concentrations are interpolated from the standard curve. Total hands-on-to-result time is approximately 3.5 hours.

Safety & Handling

For scientific research use only (RUO). This kit must not be used in diagnostic, therapeutic, or other medical applications. Handle all samples and reagents using standard laboratory safety practices.

Q.Which sample types are compatible with this kit?
A.The kit is validated for serum, plasma, and other biological fluids. Centrifuge samples to remove particulates and follow the manual's dilution guidance for your matrix.
Q.What are the detection range and sensitivity?
A.The assay quantifies HMGB1 (High mobility group protein B1) across 31.25~2000pg/mL with a sensitivity of 18.75pg/mL. Dilute samples expected to exceed the upper limit into the linear range.
Q.How long does the assay take and what format is it?
A.This is a sandwich-format ELISA that completes in approximately 3.5 hours from first incubation to reading.
Q.What instrument do I need to read the results?
A.Results are read on a standard microplate reader (colorimetric readout). Concentrations are calculated from the kit standard curve.
Q.Can this kit be used for diagnostic purposes?
A.No. It is strictly for scientific research use only (RUO) and must not be used in diagnostic or therapeutic applications.
