Scientific Background

Prolyl endopeptidase (PE) also known as prolyl oligopeptidase or post-proline cleaving enzyme is an enzyme that in humans is encoded by the PREP gene.Prolyl endopeptidase is a large cytosolic enzyme that belongs to a distinct class of serine peptidases. It was first described in the cytosol of rabbit brain as an oligopeptidase, which degrades the nonapeptide bradykinin at the Pro-Phe bond.[5] The enzyme is involved in the maturation and degradation of peptide hormones and neuropeptides such as alpha-melanocyte-stimulating hormone, luteinizing hormone-releasing hormone (LH-RH), thyrotropin-rele.

Assay Principle

This assay employs a two-site sandwich ELISA to quantitate PREP in samples. An antibody specific for PREP has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPREP present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PREP is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PREP bound in the initial step. The color development is stopped and the intensity of the color is measured.
