Scientific Background

CD64 is a type of integral membrane glycoprotein known as an Fc receptor that binds monomeric IgG-type antibodies with high affinity. It is more commonly known as Fc-gamma receptor 1 (Fc gamma RI). After binding IgG, CD64 interacts with an accessory chain known as the common gamma chain (gamma chain), which possesses an ITAM motif that is necessary for triggering cellular activation.Structurally CD64 is composed of a signal peptide that allows its transport to the surface of a cell, three extracellular immunoglobulin domains of the C2-type that it uses to bind antibody, a hydrophobic transmemb.

Assay Principle

This assay employs a two-site sandwich ELISA to quantitate FcGRI in samples. An antibody specific for FcGRI has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyFcGRI present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for FcGRI is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of FcGRI bound in the initial step. The color development is stopped and the intensity of the color is measured.
