Product Overview
JMJD2B (JmjC domain-containing histone demethylation protein 3A) is a 1,064 amino acid protein encoded by the human gene JMJD2B. JMJD2B belongs to the JMJD2B histone demethylase family and contains one JmjC domain, one JmjN domain, two PHD-type zinc fingers and two Tudor domains. The two Tudor domains recognize and bind methylated histones and have an interdigitated structure; the unusual fold is required for its ability to bind methylated histone tails. JMJD2B is a histone demethylase that specifically demethylates Lys 9 residues of Histone H3, thereby playing a role in histone code. It does not demethylate Histone H3 Lys 4, H3 Lys 27, H3 Lys 36 or H4 Lys 20, however, and is only able to demethylate trimethylated H3 Lys-9 and has weaker activity than JMJD2A, JMJD2C and JMJD2D. JMJD2B demethyl-ation of Lysine residues will generate formaldehyde and succinate. JMJD2B is a ubiquitously expressed nuclear protein.

For IHC-P, heat-induced epitope retrieval (HIER) is typically required. Recommended starting conditions: citrate buffer pH 6.0 (microwave or pressure cooker, ~20 min) for rodent samples; EDTA buffer pH 9.0 for human clinical samples. After retrieval, allow sections to cool gradually before proceeding to blocking. Include a KDM4B-positive tissue control (confirmed by literature or database expression data) and a negative isotype control in every run. Recommended dilution: IHC 1:50-1:200. Refer to the product datasheet for any target-specific retrieval guidance.
This antibody has been experimentally validated in: Human. Cross-reactivity with other species has not been systematically evaluated. If your sample species is not listed, assess immunogen sequence homology to your target species ortholog. The immunogen used is: "Synthetic peptide of human KDM4B" — higher sequence identity between species increases the probability of cross-reactivity, but experimental validation with appropriate controls is necessary before drawing scientific conclusions.

Aliquot into single-use volumes before first freeze. If working with the antibody frequently, keep a working stock at 4°C for up to 4 weeks — add 0.02% sodium azide to the working stock as a bacteriostatic agent if azide is not already present in the buffer. Monitor performance against a validated positive control when transitioning between storage aliquots or lots.
