Product Overview

MTOR, also named as FRAP1, FRAP, FRAP2 and RAPT1, belongs to the PI3/PI4-kinase family. MTOR is a Ser/Thr protein kinase that functions as an ATP and amino acid sensor to balance nutrient availability and cell growth. MTOR is Kinase subunit of both mTORC1 and mTORC2, which regulate cell growth and survival in response to nutrient and hormonal signals. mTORC1 is activated in response to growth factors or amino-acids. mTORC2 is also activated by growth factors, but seems to be nutrient-insensitive. mTORC2 seems to function upstream of Rho GTPases to regulate the actin cytoskeleton, probably by activating one or more Rho-type guanine nucleotide exchange factors. mTORC2 promotes the serum-induced formation of stress-fibers or F-actin. The antibody is specific to MTOR.

What is the recommended Western Blot dilution for the MTOR antibody?

The recommended starting dilution for Western Blot is WB 1:500-1:2000. Optimal dilution should be determined empirically for each laboratory setup, sample type, and detection system. Use freshly prepared lysates with protease inhibitors; add phosphatase inhibitors if studying phosphorylation-dependent forms of MTOR. Expected band size: Calculated 289kDa, Observed 289kDa. If background is high, increase blocking (5% non-fat milk or BSA in TBST) and/or increase wash stringency.

This antibody has been experimentally validated in: Human, Mouse, Rat. Cross-reactivity with other species has not been systematically evaluated. If your sample species is not listed, assess immunogen sequence homology to your target species ortholog. The immunogen used is: "Recombinant protein corresponding to Mouse mTOR" — higher sequence identity between species increases the probability of cross-reactivity, but experimental validation with appropriate controls is necessary before drawing scientific conclusions.
