Protein disulfide isomerase (PDI) is a thiol-disulfide oxidoreductase that facilitates protein folding and quality control in the endoplasmic reticulum (ER). In neurons, PDI plays a protective role by preventing the accumulation of misfolded proteins and maintaining redox homeostasis.
Under conditions of ER stress—common in neurodegenerative diseases—PDI expression is upregulated as part of the unfolded protein response (UPR). However, excessive or dysfunctional PDI activity has been linked to protein aggregation and oxidative damage in Alzheimer’s, ALS, and Parkinson’s disease.
PDI’s dual role in neuroprotection and pathology makes it a compelling target for therapeutic strategies aimed at modulating ER stress and proteostasis in neurodegeneration.
A 1:1000 dilution of SPC-114 was sufficient for detection of PDI in 20 µg of HeLa cell lysate by ECL immunoblot analysis.
