Overview

RBCK1 Antibody is an antibody targeting RBCK1, raised in Rabbit for protein detection and localization studies where these specifications are required.

Key elements and design rationale

  • Target: RBCK1 (reported localization: Cytoplasmic).
  • Antibody identity: Polyclonal (rabbit origin); Rabbit Ig.
  • Conjugate/label: Unconjugated (affects detection chemistry and multiplex compatibility).
  • Format: Purified.
  • Species reactivity: Human, Mouse.
  • Listed applications: WB (refer to on-page specifications for application-specific guidance).

Biological background

RanBP-type and C3HC4-type zinc finger-containing protein 1 is an E3 ubiquitin-protein ligase which accepts ubiquitin from specific E2 ubiquitin-conjugating enzymes, such as UBE2L3/UBCM4, and then transfers it to substrates. Functions as an E3 ligase for oxidized IREB2 and both heme and oxygen are necessary for IREB2 ubiquitination. Promotes ubiquitination of TAB2 and IRF3 and their degradation by the proteasome. Component of the LUBAC complex which conjugates linear ('Met-1'-linked) polyubiquitin chains to substrates and plays a key role in NF- kappa-B activation and regulation of inflammation. LUBAC conjugates linear polyubiquitin to IKBKG and RIPK1 and is involved in activation of the canonical NF-kappa-B and the JNK signaling pathways. Linear ubiquitination mediated by the LUBAC complex interferes with TNF-induced cell death and thereby prevents inflammation. LUBAC is proposed to be recruited to the TNF-R1 signaling complex (TNF-RSC) following polyubiquitination of TNF- RSC components by BIRC2 and/or BIRC3 and to conjugate linear polyubiquitin to IKBKG and possibly other components contributing to the stability of the complex. Together with FAM105B/otulin, the LUBAC complex regulates the canonical Wnt signaling during angiogenesis. Binds polyubiquitin of different linkage types. [UniProt]

Research relevance and current trends

  • Comparative expression profiling across cell types, tissues, or perturbations (e.g., drug treatment, genetic editing, or differentiation).
  • Subcellular localization and trafficking studies, including co-localization with pathway markers in microscopy-based assays.
  • Integration of protein-level measurements with transcriptomics or proteomics to relate abundance to regulation and phenotype.

Common research applications

  • Western blotting: researchers commonly compare relative signal levels across conditions and use appropriate negative/positive controls for interpretation.

Interpretation should account for antibody-dependent factors such as epitope accessibility, isoforms, and sample preparation differences across workflows.

Notes for experimental interpretation

  • Isoforms and PTMs: many targets have multiple isoforms and post-translational modifications that can shift apparent signal or localization; interpret bands/signals accordingly.
  • Epitope context: binding can depend on protein conformation and sample processing; region information in the title/immunogen can help anticipate what may be detected.
  • Species differences: predicted or validated reactivity may vary by ortholog sequence and sample context; confirm in your model system.
  • Control concepts: include negative controls (no-primary/isotype), and where possible genetic controls (KO/KD) or independent antibodies to strengthen conclusions.
