Target Thyroid Hormone Receptor

Thyroid hormone receptors (TRs) are ligand-dependent transcription factors belonging to the nuclear receptor superfamily, which also includes steroid and retinoic acid receptors. Activated by triiodothyronine (T3), TRs regulate the expression of genes involved in metabolism, growth, and cellular differentiation. In the central nervous system, TRs are essential for brain development, synaptic plasticity, and neurogenesis.

During early neurodevelopment, thyroid hormones influence neuronal migration, myelination, and axonal growth. In the adult brain, TRs continue to modulate neurophysiological processes, including mitochondrial function, oxidative stress response, and neurotransmitter signaling. Disruption of thyroid hormone signaling—whether due to receptor mutations, hormone deficiency, or altered transport—has been linked to cognitive impairment, mood disorders, and increased risk of neurodegenerative diseases.

Emerging evidence implicates TR dysfunction in the pathogenesis of Alzheimer’s disease, Parkinson’s disease, and multiple sclerosis. Aberrant TR signaling may contribute to neuroinflammation, impaired synaptic maintenance, and dysregulated energy metabolism—key features of neurodegeneration. Additionally, TRs interact with other nuclear receptors and epigenetic regulators, positioning them as integrative hubs in neuronal gene expression networks.

Given their broad regulatory scope and sensitivity to metabolic and environmental cues, thyroid hormone receptors represent promising targets for therapeutic intervention in neurodegenerative disease. Understanding their role in brain homeostasis is critical for developing strategies to preserve cognitive function and neuronal resilience.

A 1:500 dilution of SMC-561 was sufficient for detection of Pan-Thyroid hormone receptor in 10 ug of Hep G2 Human Hepatoblastoma Cell lysate by ECL immunoblot analysis using goat anti-mouse IgG:HRP as the secondary antibody.
