| Immunogen | A synthesized peptide derived from human Cytochrome P450 3A4 Cytochromes P450 are a group of heme-thiolate monooxygenases. In liver microsomes, this enzyme is involved in an NADPH-dependent electron transport pathway. It performs a variety of oxidation reactions (e.g. caffeine 8-oxidation, omeprazole sulphoxidation, midazolam 1'-hydroxylation and midazolam 4-hydroxylation) of structurally unrelated compounds, including steroids, fatty acids, and xenobiotics. The enzyme also hydroxylates etoposide. |

This description is intended to help interpret the antibody design and the biological context of the target using the fields provided in the catalog record, alongside general experimental considerations.
Biological background
CYP3A4 (protein: P2X purinoceptor 1) is a commonly studied target in molecular and cellular biology. Functional context (as provided): Induces cartilage and bone formation (PubMed:3201241). Stimulates the differentiation of myoblasts into osteoblasts via the EIF2AK3-EIF2A- ATF4 pathway. BMP2 activation of EIF2AK3 stimulates phosphorylation of EIF2A which leads to increased expression of ATF4 which plays a central role in osteoblast differentiation. In addition stimulates TMEM119, which upregulates the expression of ATF4 (PubMed:24362451). . Reported cellular localization context: Secreted. Tissue expression notes (as provided): Particularly abundant in lung, spleen and colon and in low but significant levels in heart, brain, placenta, liver, skeletal muscle, kidney, pancreas, prostate, ovary and small intestine.
Research relevance and current trends
- Research context keywords from the source record include: Cardiovascular,Cytochromes,Growth Factors/Hormones,Lipases,Lipid and Lipoprotein Metabolism,Lipid Metabolism,Lipids/Lipoproteins,Metabolic Signaling Pathways,Metabolism,Mitochondrial Metabolism,Pathways and Processes,Signal Transduction.
- Current studies often focus on connecting target abundance/localization to pathway perturbations across models, tissues, and cell states.
- Quantitative and multiplexed assays (e.g., imaging + immunoblot panels) are commonly used to compare phenotypes across conditions and time-courses.
Common research applications
- Western blotting (WB): assess relative target abundance across samples, treatments, or time-points.
- Immunohistochemistry (IHC): evaluate spatial distribution of target-positive staining in tissue architecture.
- Immunoprecipitation (IP): enrich target complexes for downstream immunoblot or interaction analyses.
Workflow ideas (metafield): Validate CYP3A4 antibody specificity using KO/KD control samples (WB/IF/IHC as appropriate), Detect CYP3A4 expression by Western blot in cell or tissue lysates, Detect CYP3A4 in FFPE tissue sections by immunohistochemistry, Enrich CYP3A4 by immunoprecipitation from lysates for downstream analysis
Notes for experimental interpretation
- Consider isoforms and post-translational modifications (PTMs) that may shift apparent molecular weight or epitope accessibility.
- Apparent molecular weight may vary by sample type and processing (observed MW: 32 kDa; calculated MW: 44702 MW).
- Control concepts: include appropriate negative controls (e.g., isotype, KO/KD samples) and orthogonal validation when feasible.
Additional product details (from the source record)
- Molecular weight (observed): 32 kDa
- Cellular localization (provided): Secreted.
- Tissue details (provided): Particularly abundant in lung, spleen and colon and in low but significant levels in heart, brain, placenta, liver, skeletal muscle, kidney, pancreas, prostate, ovary and small intestine.
