Product Overview
This rabbit polyclonal antibody targets fumarate hydratase, mitochondrial (gene FH) in human, mouse and rat samples and is validated for flow cytometry, ICC/IF, IHC and Western blot. The predicted molecular weight is 54.6 kDa, and the supplier reports an observed band at about 49 kDa.
Fumarate hydratase (FH, fumarase) is a tricarboxylic acid (TCA) cycle enzyme in mitochondria that converts fumarate to L-malate; a cytosolic form also exists. Loss of FH causes fumarate to build up, and inherited FH mutations cause hereditary leiomyomatosis and renal cell cancer, so FH is studied in cancer metabolism and mitochondrial function.

Samples with a confirmed band (WB): rat lymphaden tissue, rat small intestine tissue, rat gaster tissue, mouse kidney tissue, mouse testis tissue, mouse gaster tissue, human K562 cells, human U937 cells, human colon cancer tissue, human lung cancer tissue, human placenta tissue, mouse small intestine tissue, human mammary cancer tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with confirmed staining (ICC/IF): PC-3 cells.
Samples with a confirmed signal (flow cytometry): A431 cells, PC-3 cells.
Conditions in the example images (IHC): heat-mediated antigen retrieval in citrate buffer (pH 6).
