Target GPER1

Overview

Anti-GPER1/GPR30 (extracellular)-FITC Antibody is an antibody targeting G Protein-Coupled Estrogen Receptor 1, G Protein-Coupled Receptor 30, Membrane Estrogen Receptor, Chemoattractant Receptor-Like 2, GPER Polyclonal raised in Rabbit (Fluorescein isothiocyanate (FITC)). This antibody is commonly used in FC to detect, localize, or compare expression of the target across samples.

Key elements and design rationale

  • Target: G Protein-Coupled Estrogen Receptor 1, G Protein-Coupled Receptor 30, Membrane Estrogen Receptor, Chemoattractant Receptor-Like 2, GPER (also reported as G Protein-Coupled Estrogen Receptor 1, G Protein-Coupled Receptor 30, Membrane Estrogen Receptor, Chemoattractant Receptor-Like 2, GPER).
  • Immunogen/epitope region: 3rd extracellular loop.
  • Homology note: Rat - identical; human - 13/14 amino acid residues identical (informative for cross-species interpretation).
  • Species reactivity (as provided): Human, Rat, Mouse.
  • Lot quality control (as provided): Western blot analysis (unlabeled antibody, #AER-049), and direct flow cytometry (labeled antibody)..
  • Peptide confirmation: Confirmed by amino acid analysis and mass spectrometry.
  • Blocking peptide: Available for antigen preadsorption control where appropriate.
  • Conjugate/format: Fluorescein isothiocyanate (FITC) (may affect detection channel and background).

These attributes help researchers interpret whether signal reflects the intended target in a given assay and sample context.

Biological background

GPER1/GPR30 is an estrogen receptor belonging to the superfamily of G-protein coupled receptors (GPCRs). Like all members, GPER1 has seven membrane-spanning domains, an extracellular N-terminus and a cytoplasmic C-terminal domain. It is widely expressed in the central nervous system (CNS) and it binds 17β-estradiol (17β-E) with high affinity1.Estrogens have long been known as important regulators of the female reproductive functions, however, is now commonly accepted that estrogens and androgens have important functions in both female and male physiology and pathology. As opposed to the epigenetic regulatory role in estrogen signaling carried out by ER-α and ER-β estrogen receptors, signaling from GPER1 is responsible for the rapid non-genomic actions of estrogens that activate various protein-kinase cascades1.

Research relevance and current trends

  • Comparing target expression across perturbations, genotypes, or treatment conditions.
  • Interpreting localization shifts alongside pathway or phenotypic readouts.
  • Using orthogonal controls (KO/KD, peptide competition, isotype concepts) to support conclusions.

Common research applications

  • Flow cytometry (direct/indirect): quantify target-positive populations and shifts in expression across subsets.

Interpretation typically benefits from comparing matched sample sets (e.g., treated vs control, WT vs KO/KD) and using orthogonal readouts where feasible.

Notes for experimental interpretation

  • Isoforms and post-translational modifications can shift apparent molecular weight or epitope accessibility across samples.
  • Cross-species signal may depend on epitope conservation; consult the provided homology note when selecting models.
  • Permeabilization, fixation, and antigen retrieval can change accessibility of intracellular vs extracellular epitopes.
  • Conceptual control: antigen preadsorption (blocking peptide) can help assess signal dependence on the immunogen region.
  • Provided control suggestions: Negative control: RIC-001-F.
  • Application notes: see product-specific dilution/usage notes and control concepts provided in the dataset.
