Product Overview
This rabbit polyclonal antibody recognizes histone deacetylase 4 (gene HDAC4) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF and immunoprecipitation. The predicted molecular weight is 119.0 kDa, and the supplier reports an observed band at about 140 kDa.
Histone deacetylase 4 (HDAC4) is a class IIa deacetylase that shuttles between the nucleus and cytoplasm in response to phosphorylation. In the nucleus it binds MEF2 transcription factors and represses their target genes, influencing muscle and bone development and neuronal function. Class IIa HDACs have weak intrinsic catalytic activity and recruit other corepressors, and HDAC4 is studied in muscle biology and chondrocyte maturation.

Samples with a confirmed band (WB): human Jurkat cells, human THP-1 cells, human HepG2 cells, human SH-SY5Y cells, rat brain tissue, mouse brain tissue, mouse NIH3T3 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 8% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human cerebral cortex tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): Jurkat cells.
Immunogen
Recombinant human HDAC4 fragment (Gln4–Gln213), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 98.6% and 98.1%, respectively.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places histone deacetylase 4 in the nucleus and cytoplasm. Tissue expression noted by UniProt (human): Ubiquitous. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
