Product Overview
This rabbit polyclonal antibody targets histone deacetylase 9 (gene HDAC9) in human, mouse and rat samples and is validated for Western blot, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 111.3 kDa, and the supplier reports an observed band at about 150 kDa.
Histone deacetylase 9 (HDAC9) is a class IIa deacetylase that represses MEF2-dependent transcription. A short splice form known as MITR lacks the deacetylase domain but still acts as a repressor by recruiting other corepressors. HDAC9 is studied in muscle and heart gene regulation, regulatory T-cell function and vascular disease.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human, Mouse, Rat) |
|---|---|
| Immunocytochemistry / Immunofluorescence | 4 µg/mL (Human) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): rat testis tissue, rat liver tissue, mouse testis tissue, mouse liver tissue, mouse RAW264.7 cells, human THP-1 cells, human HL-60 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with confirmed staining (ICC/IF): HeLa cells.
Samples with a confirmed signal (flow cytometry): THP-1 cells.
Immunogen
Recombinant human HDAC9 fragment (Ser7–Gln469), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places histone deacetylase 9 in the nucleus. Tissue expression noted by UniProt (human): Broadly expressed, with highest levels in brain, heart, muscle and testis. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
