| Immunogen | A synthesized peptide derived from human IKKi/IKKe Phosphorylates inhibitors of NF-kappa-B thus leading to the dissociation of the inhibitor/NF-kappa-B complex and ultimately the degradation of the inhibitor. May play a special role in the immune response. Protects cells against DNA damage-induced cell death. |

Product Overview
Clone AFAO-9 is a rabbit monoclonal antibody that recognizes inhibitor of nuclear factor kappa-B kinase subunit epsilon (gene IKBKE) in human samples and is validated for Western blot, IHC, ICC/IF and flow cytometry. The predicted molecular weight is 80.5 kDa, and the supplier reports an observed band at about 80 kDa.
IKKε (IKBKE, also called IKKi) is a non-canonical IκB kinase related to TBK1. After viral RNA or DNA sensing, it phosphorylates IRF3 and IRF7 to switch on type I interferon genes, and it also contributes to NF-κB and STAT signaling, linking it to antiviral immunity and inflammation.

Samples with a confirmed band (WB): human Raji cells, human Daudi cells, human Ramos cells, human HeLa cells, human THP-1 cells, human RT4 cells, mouse thymus tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Immunogen
Synthetic peptide based on the sequence of human IKKi/IKKe.
Reactivity Notes
The supplier lists reactivity with human. UniProt places inhibitor of nuclear factor kappa-B kinase subunit epsilon in the cytoplasm and nucleus. Tissue expression noted by UniProt (human): Highly expressed in spleen followed by thymus, peripheral blood leukocytes, pancreas, placenta. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
