Anti-KCNN3 (KCa2.3, SK3) (N-term) Antibody (#APC-025) is a highly specific antibody directed against an epitope of the human protein. The antibody can be used in western blot, immunocytochemistry, and immunohistochemistry applications. It has been designed to recognize KCNN3 from human, rat, and mouse samples. Anti-KCNN3 (KCa2.3, SK3) (N-term)-ATTO Fluor-594 Antibody (#APC-025-AR) is directly labeled with an ATTO-594 fluorescent dye. ATTO dyes are characterized by strong absorption (high extinction coefficient), high fluorescence quantum yield, and high photo-stability. The ATTO-594 fluorescent label belongs to the class of Rhodamine dyes and can be used with fluorescent equipment typically optimized to detect Texas Red and Alexa-594. Anti-KCNN3 (KCa2.3, SK3) (N-term)-ATTO Fluor-594 Antibody is specially suited to experiments requiring simultaneous labeling of different markers.

Target KCNN3

Overview

Anti-KCNN3 (KCa2.3, SK3) (N-term)-ATTO Fluor-594 Antibody is an antibody targeting Small conductance calcium-activated potassium channel protein 3, SKCa3 Polyclonal raised in Rabbit (ATTO-594. Maximum absorption 601 nm; Maximum fluorescence 627 nm. The fluorescence is excited most efficiently in the 580 - 615 nm range. This label belongs to the class of Rhodamine dyes and can be used with fluorescent equipment typically optimized to detect Texas Red and Alexa-594.). This antibody is commonly used in FC, IC, IF, IHC to detect, localize, or compare expression of the target across samples.

Key elements and design rationale

  • Target: Small conductance calcium-activated potassium channel protein 3, SKCa3 (also reported as Small conductance calcium-activated potassium channel protein 3, SKCa3).
  • Immunogen/epitope region: Intracellular, N-terminus.
  • Homology note: Rat, mouse, pig - identical (informative for cross-species interpretation).
  • Species reactivity (as provided): Human, Rat, Mouse.
  • KO-validated: yes (validation context may be assay-dependent).
  • Cited use: ICC, FC (literature use does not guarantee performance in every setup).
  • Lot quality control (as provided): Western blot analysis (unlabeled antibody, #APC-025), and immunohistochemistry (labeled antibody)..
  • Peptide confirmation: Confirmed by amino acid analysis and mass spectrometry.

These attributes help researchers interpret whether signal reflects the intended target in a given assay and sample context.

Biological background

KCa2.3 (KCNN3, SK3) is a member of the Ca2+-activated K+ channel family with small conductance that includes KCa2.1 (KCNN1, SK1) and KCa2.2 (KCNN2, SK2). The channel is voltage insensitive and is activated by intracellular Ca2+ in the submicromolar range. It has, though, a similar topology to that of voltage-dependent K+ channels (KV channels), that is six transmembrane domains and intracellular N- and C-termini.

Research relevance and current trends

  • Linking transporter/channel abundance to ionic homeostasis and excitability-related phenotypes.
  • Studying compartment-specific localization (surface vs intracellular pools) and trafficking dynamics.
  • Combining antibody readouts with functional assays for more complete interpretation.

Common research applications

  • Immunohistochemistry (IHC): examine spatial distribution in tissue and relate signal to cell-type composition.
  • Immunofluorescence/ICC: assess subcellular localization and co-localization with markers in cells or sections.
  • Flow cytometry (direct/indirect): quantify target-positive populations and shifts in expression across subsets.

Interpretation typically benefits from comparing matched sample sets (e.g., treated vs control, WT vs KO/KD) and using orthogonal readouts where feasible.

Notes for experimental interpretation

  • Isoforms and post-translational modifications can shift apparent molecular weight or epitope accessibility across samples.
  • Cross-species signal may depend on epitope conservation; consult the provided homology note when selecting models.
  • Permeabilization, fixation, and antigen retrieval can change accessibility of intracellular vs extracellular epitopes.
  • Conceptual control: antigen preadsorption (blocking peptide) can help assess signal dependence on the immunogen region.
  • Conceptual control: KO/KD samples provide orthogonal support for target assignment when available.
  • Provided control suggestions: Negative control: BLP-PC025.
  • Application notes: see product-specific dilution/usage notes and control concepts provided in the dataset.

Application abbreviations: CBE- Cell-based ELISA, FC- Flow cytometry, ICC- Immunocytochemistry, IE- Indirect ELISA, IF- Immunofluorescence, IFC- Indirect flow cytometry, IHC- Immunohistochemistry, IP- Immunoprecipitation, LCI- Live cell imaging, N- Neutralization, WB- Western blot.
