Anti-Kir7.1 (extracellular) Antibody (#APC-125) is a highly specific antibody directed against an extracellular epitope of the rat protein. The antibody can be used in western blot, immunohistochemistry and indirect flow cytometry applications. It has been designed to recognize Kir7.1 from rat, mouse and human samples. Anti-Kir7.1 (extracellular)-ATTO Fluor-488 Antibody (#APC-125-AG) is directly labeled with an ATTO-488 fluorescent dye. ATTO dyes are characterized by strong absorption (high extinction coefficient), high fluorescence quantum yield, and high photo-stability. The ATTO-488 label is analogous to the well known dye fluorescein isothiocyanate (FITC) and can be used with filters typically used to detect FITC. Anti-Kir7.1 (extracellular)-ATTO Fluor-488 Antibody is especially suited for experiments requiring simultaneous labeling of different markers.

Target KCNJ13

Overview

Anti-Kir7.1 (extracellular)-ATTO Fluor-488 Antibody is an antibody targeting Inward rectifier potassium channel 13, KCNJ13 Polyclonal raised in Rabbit (ATTO-488. Maximum absorption 501 nm; maximum fluorescence 523 nm. The fluorescence is excited most efficiently in the 480 - 515 nm range. This label is analogous to the dye fluorescein isothiocyanate (FITC) and can be used with filters used to detect FITC.). This antibody is commonly used in IF, IHC to detect, localize, or compare expression of the target across samples.

Key elements and design rationale

  • Target: Inward rectifier potassium channel 13, KCNJ13 (also reported as Inward rectifier potassium channel 13, KCNJ13).
  • Immunogen/epitope region: Extracellular loop.
  • Homology note: Mouse - identical; human - 13/15 amino acid residues identical (informative for cross-species interpretation).
  • Species reactivity (as provided): Human, Rat, Mouse.
  • Cited use: IHC (literature use does not guarantee performance in every setup).
  • Lot quality control (as provided): Western blot analysis (unlabeled antibody, #APC-125), and immunohistochemistry (labeled antibody)..
  • Peptide confirmation: Confirmed by amino acid analysis and mass spectrometry.
  • Blocking peptide: Available for antigen preadsorption control where appropriate.

These attributes help researchers interpret whether signal reflects the intended target in a given assay and sample context.

Biological background

Kir7.1 (KCNJ13) is a member of the inward rectifying K+ channel family. The family includes 15 members that are structurally and functionally different from the voltage-dependent K+ channels. The family's protein topology consists of two transmembrane domains that flank a single and highly conserved pore region with intracellular N- and C-termini. As is the case for the voltage-dependent K+ channels the functional unit for the Kir channels is composed of four subunits that can assemble as either homo- or heteromers.Kir channels are characterized by a K+ efflux that is limited by depolarizing membrane potentials thus making them essential for controlling resting membrane potential and K+ homeostasis1.Kir7.1, an inwardly rectifying K+ channel with unusual permeation properties is localized in epithelial cells of the thyroid, small intestine, kidney tubules, choroid plexus and in retinal pigment epithelium (RPE), where it forms a major component of the apical membrane K+ conductance2.A mutation in the gene encoding the channel was found to cause snowflake vitreoretinal degeneration (SVD) which is a developmental and progressive hereditary eye disorder that affects multiple tissues within the eye3.

Research relevance and current trends

  • Linking transporter/channel abundance to ionic homeostasis and excitability-related phenotypes.
  • Studying compartment-specific localization (surface vs intracellular pools) and trafficking dynamics.
  • Combining antibody readouts with functional assays for more complete interpretation.

Common research applications

  • Immunohistochemistry (IHC): examine spatial distribution in tissue and relate signal to cell-type composition.
  • Immunofluorescence/ICC: assess subcellular localization and co-localization with markers in cells or sections.

Interpretation typically benefits from comparing matched sample sets (e.g., treated vs control, WT vs KO/KD) and using orthogonal readouts where feasible.

Notes for experimental interpretation

  • Isoforms and post-translational modifications can shift apparent molecular weight or epitope accessibility across samples.
  • Cross-species signal may depend on epitope conservation; consult the provided homology note when selecting models.
  • Permeabilization, fixation, and antigen retrieval can change accessibility of intracellular vs extracellular epitopes.
  • Conceptual control: antigen preadsorption (blocking peptide) can help assess signal dependence on the immunogen region.
  • Provided control suggestions: Negative control: RIC-001-AG.
  • Application notes: see product-specific dilution/usage notes and control concepts provided in the dataset.
