Anti-KV1.1 (KCNA1) (extracellular) Antibody (#APC-161) is a highly specific antibody directed against an epitope of the rat protein. The antibody can be used in western blot, immunohistochemistry, indirect flow cytometry and live cell imaging applications. It has been designed to recognize KV1.1 from human, rat and mouse samples. Anti-KV1.1 (KCNA1) (extracellular)-ATTO Fluor-594 Antibody (#APC-161-AR) is directly labeled with an ATTO-594 fluorescent dye. ATTO dyes are characterized by strong absorption (high extinction coefficient), high fluorescence quantum yield, and high photo-stability. The ATTO-594 fluorescent label belongs to the class of Rhodamine dyes and can be used with fluorescent equipment typically optimized to detect Texas Red and Alexa-594. Anti-KV1.1 (KCNA1) (extracellular)-ATTO Fluor-594 Antibody is especially suited for experiments requiring simultaneous labeling of different markers.

Overview

Anti-KV1.1 (KCNA1) (extracellular)-ATTO Fluor-594 Antibody is an antibody targeting Potassium voltage-gated channel subfamily A member 1 Polyclonal raised in Rabbit (ATTO-594. Maximum absorption 601 nm; Maximum fluorescence 627 nm. The fluorescence is excited most efficiently in the 580 - 615 nm range. This label belongs to the class of Rhodamine dyes and can be used with fluorescent equipment typically optimized to detect Texas Red and Alexa-594.). This antibody is commonly used in IC, IF, IHC, LCI to detect, localize, or compare expression of the target across samples.

Key elements and design rationale

  • Target: Potassium voltage-gated channel subfamily A member 1 (also reported as Potassium voltage-gated channel subfamily A member 1).
  • Immunogen/epitope region: 1st extracellular loop.
  • Homology note: Mouse - identical; human - 13/14 amino acid residues identical (informative for cross-species interpretation).
  • Species reactivity (as provided): Rat, Mouse.
  • Lot quality control (as provided): Western blot analysis (unlabeled antibody, #APC-161), and immunocytochemistry (labeled antibody)..
  • Peptide confirmation: Confirmed by amino acid analysis and mass spectrometry.
  • Blocking peptide: Available for antigen preadsorption control where appropriate.
  • Conjugate/format: ATTO-594. Maximum absorption 601 nm; Maximum fluorescence 627 nm. The fluorescence is excited most efficiently in the 580 - 615 nm range. This label belongs to the class of Rhodamine dyes and can be used with fluorescent equipment typically optimized to detect Texas Red and Alexa-594. (may affect detection channel and background).

These attributes help researchers interpret whether signal reflects the intended target in a given assay and sample context.

Biological background

KV1.1 is a mammalian voltage-dependent K+ channel, homologous to the Drosophila Shaker K+ channel. KV1.1 was the first mammalian KV channel to be cloned from mouse brain.1 Eight Shaker-related genes exist in mammals constituting the KV1, subfamily of the large KV channel family of genes.2A functional KV1 channel is either a membrane spanning homotetramer or heterotetramer, which is composed of members of the same subfamily. In addition several auxiliary subunits and intracellular proteins might interact with the channel and affect its function.

Research relevance and current trends

  • Mapping receptor/channel localization across neuronal subtypes and subcellular compartments.
  • Linking trafficking or surface expression changes to activity-dependent signaling and plasticity.
  • Using KO/KD or blocking-peptide concepts to strengthen antibody-based target assignment.

Common research applications

  • Immunohistochemistry (IHC): examine spatial distribution in tissue and relate signal to cell-type composition.
  • Immunofluorescence/ICC: assess subcellular localization and co-localization with markers in cells or sections.
  • Live cell imaging (LCI): support extracellular-epitope detection on non-permeabilized cells when appropriate.

Interpretation typically benefits from comparing matched sample sets (e.g., treated vs control, WT vs KO/KD) and using orthogonal readouts where feasible.

Notes for experimental interpretation

  • Isoforms and post-translational modifications can shift apparent molecular weight or epitope accessibility across samples.
  • Cross-species signal may depend on epitope conservation; consult the provided homology note when selecting models.
  • Permeabilization, fixation, and antigen retrieval can change accessibility of intracellular vs extracellular epitopes.
  • Conceptual control: antigen preadsorption (blocking peptide) can help assess signal dependence on the immunogen region.
  • Provided control suggestions: Negative control: BLP-PC161.
  • Application notes: see product-specific dilution/usage notes and control concepts provided in the dataset.
