Overview
Anti-LRRC26 Antibody is an antibody targeting Leucine-rich repeat-containing protein 26, BK channel auxiliary gamma subunit LRRC26, Cytokeratin-associated protein in cancer, CAPC Polyclonal raised in Rabbit (Unconjugated). This antibody is commonly used in WB to detect, localize, or compare expression of the target across samples.
Key elements and design rationale
- Target: Leucine-rich repeat-containing protein 26, BK channel auxiliary gamma subunit LRRC26, Cytokeratin-associated protein in cancer, CAPC (also reported as Leucine-rich repeat-containing protein 26, BK channel auxiliary gamma subunit LRRC26, Cytokeratin-associated protein in cancer, CAPC).
- Immunogen/epitope region: Intracellular, C-terminus.
- Homology note: Mouse - 12/14 amino acid residues identical (informative for cross-species interpretation).
- Species reactivity (as provided): Rat, Mouse.
- Specificity statement (as provided): Won’t recognize human LRRC26..
- Lot quality control (as provided): Western blot analysis.
- Peptide confirmation: Confirmed by amino acid analysis and mass spectrometry.
- Blocking peptide: Available for antigen preadsorption control where appropriate.
These attributes help researchers interpret whether signal reflects the intended target in a given assay and sample context.
Biological background
BK (KCa1.1) channels are critically involved in diverse physiological processes in both electrically excitable and non-excitable cells.Leucine-rich repeat containing protein 26 (also known as BK channel auxiliary gamma subunit or LRRC26) is a single spanning transmembrane protein of ~37 kDa. LRRC26 protein contains five major leucine rich repeats in its extracellular domain which are all required for modulating BK channel activity. The single transmembrane segment of LRRC26 is required for co-assembly with the BKα subunit complex. When LRRC26 is acutely knocked down in LNCaP cells, the voltage-dependent activation of endogenous BK current shifts from a low to a high V1/2 value, whereas recombinant expression of LRRC26 cDNA in naïve cells (such as PC3 prostate cancer cells and HEK 293 cells) normally lacking LRRC26 shifts BK channel activation from high to low V1/2 values, reproducing the channel behavior observed in LNCaP cells.
Research relevance and current trends
- Linking transporter/channel abundance to ionic homeostasis and excitability-related phenotypes.
- Studying compartment-specific localization (surface vs intracellular pools) and trafficking dynamics.
- Combining antibody readouts with functional assays for more complete interpretation.
Common research applications
- Western blot (WB): compare target abundance/size across lysates and conditions; consider isoforms/PTMs.
Interpretation typically benefits from comparing matched sample sets (e.g., treated vs control, WT vs KO/KD) and using orthogonal readouts where feasible.
Notes for experimental interpretation
- Isoforms and post-translational modifications can shift apparent molecular weight or epitope accessibility across samples.
- Cross-species signal may depend on epitope conservation; consult the provided homology note when selecting models.
- Permeabilization, fixation, and antigen retrieval can change accessibility of intracellular vs extracellular epitopes.
- Conceptual control: antigen preadsorption (blocking peptide) can help assess signal dependence on the immunogen region.
- Provided control suggestions: Negative control: BLP-PC070.
- Application notes: see product-specific dilution/usage notes and control concepts provided in the dataset.
