This description is intended to help interpret the antibody design and the biological context of the target using the fields provided in the catalog record, alongside general experimental considerations.

Biological background

MFN2 (protein: T-cell surface glycoprotein CD4) is a commonly studied target in molecular and cellular biology. Functional context (as provided): Essential transmembrane GTPase, which mediates mitochondrial fusion. Fusion of mitochondria occurs in many cell types and constitutes an important step in mitochondria morphology, which is balanced between fusion and fission. MFN2 acts independently of the cytoskeleton. It therefore plays a central role in mitochondrial metabolism and may be associated with obesity and/or apoptosis processes. Overexpression induces the formation of mitochondrial networks. Plays an important role in the regulation of vascular smooth muscle cell proliferation. Involved in the clearance of damaged mitochondria via selective autophagy (mitophagy). Is required for PARK2 recruitment to dysfunctional mitochondria. Involved in the control of unfolded protein response (UPR) upon ER stress including activation of apoptosis and autophagy during ER stress. Acts as an upstream regulator of EIF2AK3 and suppresses EIF2AK3 activation under basal conditions. . Reported cellular localization context: Mitochondrion outer membrane ; Multi- pass membrane protein . Colocalizes with BAX during apoptosis. Tissue expression notes (as provided): Ubiquitous; expressed at low level. Highly expressed in heart and kidney. .

Research relevance and current trends

  • Research context keywords from the source record include: Mitochondrial Markers,Mitochondrial Metabolism,Neurodegenerative Disease,Neurology Process,Neuroscience,Organelles,Pathways and Processes,Signal Transduction,Subcellular Markers,Tags & Cell Markers.
  • Current studies often focus on connecting target abundance/localization to pathway perturbations across models, tissues, and cell states.
  • Quantitative and multiplexed assays (e.g., imaging + immunoblot panels) are commonly used to compare phenotypes across conditions and time-courses.

Common research applications

  • Western blotting (WB): assess relative target abundance across samples, treatments, or time-points.
  • Immunohistochemistry (IHC): evaluate spatial distribution of target-positive staining in tissue architecture.
  • Immunofluorescence/ICC (IF/ICC): visualize subcellular localization patterns and cell-to-cell heterogeneity.

Workflow ideas (metafield): Validate MFN2 antibody specificity using KO/KD control samples (WB/IF/IHC as appropriate), Detect MFN2 expression by Western blot in cell or tissue lysates, Detect MFN2 in FFPE tissue sections by immunohistochemistry, Localize MFN2 by immunofluorescence/immunocytochemistry in cultured cells

Notes for experimental interpretation

  • Consider isoforms and post-translational modifications (PTMs) that may shift apparent molecular weight or epitope accessibility.
  • Apparent molecular weight may vary by sample type and processing (observed MW: 80 kDa; calculated MW: 86402 MW).
  • Control concepts: include appropriate negative controls (e.g., isotype, KO/KD samples) and orthogonal validation when feasible.

Additional product details (from the source record)

  • Molecular weight (observed): 80 kDa
  • Cellular localization (provided): Mitochondrion outer membrane ; Multi- pass membrane protein . Colocalizes with BAX during apoptosis.
  • Tissue details (provided): Ubiquitous; expressed at low level. Highly expressed in heart and kidney. .

My colleagues were satisfied with the WB result of your anti-Mitofusin 2 Rabbit Monoclonal antibody. However we have observed positive staining in testis mitochondrion outer membrane using this antibody. Is that expected? Could you tell me where is MFN2 supposed to be expressed?
Based on literature, testis does express MFN2. Generally MFN2 expresses in mitochondrion outer membrane. Regarding which tissues have MFN2 expression, here are a few articles citing expression in various tissues: Bone marrow, Pubmed ID: 9039502 Brain, Pubmed ID: 14702039 Pancreas, Pubmed ID: 15489334 Testis, Pubmed ID: 17974005
We are currently using anti-Mitofusin 2 Rabbit Monoclonal antibody M00461 for rat tissue, and we are content with the WB results. The species of reactivity given in the datasheet says human, mouse, rat. Is it true that the antibody can work on monkey tissues as well?
The anti-Mitofusin 2 Rabbit Monoclonal antibody (M00461) has not been tested for cross reactivity specifically with monkey tissues, though there is a good chance of cross reactivity. We have an innovator award program that if you test this antibody and show it works in monkey you can get your next antibody for free. Please contact me if I can help you with anything.
