Key elements and design rationale

  • Target: OCLN (Occludin).
  • Antibody format: Monoclonal; clone IEH-15; isotype Rabbit IgG.
  • Host: Rabbit.
  • Species reactivity: Human,Mouse,Rat (confirm in your model system with appropriate controls).

This description is intended to help interpret the antibody design and the biological context of the target using the fields provided in the catalog record, alongside general experimental considerations.

Biological background

OCLN (protein: Glycogen synthase kinase-3 beta (gsk3b)) is a commonly studied target in molecular and cellular biology. Functional context (as provided): May play a role in the formation and regulation of the tight junction (TJ) paracellular permeability barrier. It is able to induce adhesion when expressed in cells lacking tight junctions. . Reported cellular localization context: Membrane; Multi-pass membrane protein. Cell junction, tight junction. Tissue expression notes (as provided): Localized at tight junctions of both epithelial and endothelial cells. Highly expressed in kidney. Not detected in testis.

Research relevance and current trends

  • Research context keywords from the source record include: Cell Adhesion,Cytoskeleton/ECM,Signal Transduction,Tight Junctions.
  • Current studies often focus on connecting target abundance/localization to pathway perturbations across models, tissues, and cell states.
  • Quantitative and multiplexed assays (e.g., imaging + immunoblot panels) are commonly used to compare phenotypes across conditions and time-courses.

Common research applications

  • Western blotting (WB): assess relative target abundance across samples, treatments, or time-points.
  • Flow cytometry: quantify target-positive populations and compare shifts in marker distributions.
  • Immunoprecipitation (IP): enrich target complexes for downstream immunoblot or interaction analyses.

Workflow ideas (metafield): Validate OCLN antibody specificity using KO/KD control samples (WB/IF/IHC as appropriate), Detect OCLN expression by Western blot in cell or tissue lysates, Quantify OCLN-positive cells by flow cytometry in single-cell suspensions, Enrich OCLN by immunoprecipitation from lysates for downstream analysis

Notes for experimental interpretation

  • Consider isoforms and post-translational modifications (PTMs) that may shift apparent molecular weight or epitope accessibility.
  • Apparent molecular weight may vary by sample type and processing (observed MW: 80 kDa; calculated MW: 59144 MW).
  • Control concepts: include appropriate negative controls (e.g., isotype, KO/KD samples) and orthogonal validation when feasible.

Additional product details (from the source record)

  • Molecular weight (observed): 80 kDa
  • Cellular localization (provided): Membrane; Multi-pass membrane protein. Cell junction, tight junction.
  • Tissue details (provided): Localized at tight junctions of both epithelial and endothelial cells. Highly expressed in kidney. Not detected in testis.

I was wanting to use your anti-Occludin Rabbit Monoclonal antibody for IP for human colon carcinoma on frozen tissues, but I want to know if it has been tested for this particular application. Has this antibody been tested and is this antibody a good choice for human colon carcinoma identification?
It shows on the product datasheet, M01246 anti-Occludin Rabbit Monoclonal antibody has been tested for IP, WB on human, mouse, rat tissues. We have an innovator award program that if you test this antibody and show it works in human colon carcinoma in IHC-frozen, you can get your next antibody for free.
Does M01246 anti-Occludin Rabbit Monoclonal antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?
As indicated on the product datasheet, M01246 anti-Occludin Rabbit Monoclonal antibody as been validated on IP. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
