Anti-P2Y12 Receptor (extracellular) Antibody (#APR-020) is a highly specific antibody directed against an extracellular epitope of the human P2RY12. The antibody can be used in western blot, live cell imaging and indirect flow cytometry applications. It has been designed to recognize P2RY12 from human, rat and mouse samples. Anti-P2Y12 Receptor (extracellular)-mFluor™ Violet 450 Antibody (#APR-020-V) is directly conjugated to mFluor™ Violet 450 fluorophore. This conjugated antibody has been developed to be used in immunofluorescent applications such as direct flow cytometry and live cell imaging. mFluor™ Violet 450 dye has a maximum absorption of 405 nm and maximum emission of 445 nm. These spectral characteristics make it an excellent replacement for Pacific Blue™ or Brilliant Violet™ 421 dyes. For flow cytometry applications, the labeled antibody can be detected using the 405 nm laser lane and a filter set of 450/50.

Target P2RY12

Overview

Anti-P2Y12 Receptor (extracellular)-mFluor™ Violet 450 Antibody is an antibody targeting P2Y purinoceptor 12, P2RY12, P2Y receptor Polyclonal raised in Rabbit (mFluor™ Violet 450. Maximum absorption 405 nm and maximum emission 445 nm. These spectral characteristics make it an excellent replacement for Pacific Blue™ or Brilliant Violet™ 421 dyes. For flow cytometry applications, the labeled antibody can be detected using the 405 nm laser lane and a filter set of 450/50.). This antibody is commonly used in FC, LCI to detect, localize, or compare expression of the target across samples.

Key elements and design rationale

  • Target: P2Y purinoceptor 12, P2RY12, P2Y receptor (also reported as P2Y purinoceptor 12, P2RY12, ADP-glucose receptor, Platelet ADP receptor, HORK3).
  • Immunogen/epitope region: 3rd extracellular loop.
  • Homology note: Rat, mouse - 12/13 amino acid residues identical. (informative for cross-species interpretation).
  • Species reactivity (as provided): Human, Rat, Mouse.
  • Lot quality control (as provided): Western blot analysis (unlabeled antibody, #APR-020), and direct flow cytometry (labeled antibody)..
  • Peptide confirmation: Confirmed by amino acid analysis and mass spectrometry.
  • Blocking peptide: Available for antigen preadsorption control where appropriate.
  • Conjugate/format: mFluor™ Violet 450. Maximum absorption 405 nm and maximum emission 445 nm. These spectral characteristics make it an excellent replacement for Pacific Blue™ or Brilliant Violet™ 421 dyes. For flow cytometry applications, the labeled antibody can be detected using the 405 nm laser lane and a filter set of 450/50. (may affect detection channel and background).

These attributes help researchers interpret whether signal reflects the intended target in a given assay and sample context.

Biological background

The P2Y receptors belong to the G-protein coupled receptor superfamily. They mediate the actions of the extracellular nucleotides (ATP, ADP, UTP and UDPA). Eight functional mammalian P2Y receptors have been described: P2Y1, P2Y2, P2Y4, P2Y6, P2Y11, P2Y12, P2Y13, and the UDP-glucose receptor, now renamed P2Y141-3.

Research relevance and current trends

  • Comparing target expression across perturbations, genotypes, or treatment conditions.
  • Interpreting localization shifts alongside pathway or phenotypic readouts.
  • Using orthogonal controls (KO/KD, peptide competition, isotype concepts) to support conclusions.

Common research applications

  • Flow cytometry (direct/indirect): quantify target-positive populations and shifts in expression across subsets.
  • Live cell imaging (LCI): support extracellular-epitope detection on non-permeabilized cells when appropriate.

Interpretation typically benefits from comparing matched sample sets (e.g., treated vs control, WT vs KO/KD) and using orthogonal readouts where feasible.

Notes for experimental interpretation

  • Isoforms and post-translational modifications can shift apparent molecular weight or epitope accessibility across samples.
  • Cross-species signal may depend on epitope conservation; consult the provided homology note when selecting models.
  • Permeabilization, fixation, and antigen retrieval can change accessibility of intracellular vs extracellular epitopes.
  • Conceptual control: antigen preadsorption (blocking peptide) can help assess signal dependence on the immunogen region.
  • Provided control suggestions: Negative control: RIC-001-V.

Application abbreviations: CBE- Cell-based ELISA, FC- Flow cytometry, ICC- Immunocytochemistry, IE- Indirect ELISA, IF- Immunofluorescence, IFC- Indirect flow cytometry, IHC- Immunohistochemistry, IP- Immunoprecipitation, LCI- Live cell imaging, N- Neutralization, WB- Western blot. Species abbreviations: H- Human, M- Mouse, R- Rat.

Recommended controls: Blocking peptide: BLP-PR020; Negative control: RIC-001-V (name: Rabbit IgG Isotype Control-mFluor™ Violet 450).
