Product Overview
This rabbit polyclonal antibody targets 3-phosphoinositide-dependent protein kinase 1 (gene PDPK1) in human, mouse and rat samples and is validated for IHC and Western blot. The predicted molecular weight is 63.2 kDa, and the supplier reports an observed band at about 70 kDa.
PDPK1 (3-phosphoinositide-dependent protein kinase 1, often called PDK1) is a master kinase of the AGC family. It phosphorylates the activation loop of AKT, S6K, RSK, SGK, PKC isoforms and other kinases, so it is central to insulin and growth factor signaling through PI3K, controlling cell growth, survival and metabolism, and is widely studied in cancer and metabolic research.

Samples with a confirmed band (WB): rat liver tissue, rat lung tissue, mouse liver tissue, mouse lung tissue, COLO320 cells, MCF-7 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with confirmed staining (IHC): mouse intestine tissue, rat testis tissue, human lung cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in citrate buffer (pH 6).
Samples with confirmed staining (IHC): human placenta tissue, mouse small intestine tissue.

Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places 3-phosphoinositide-dependent protein kinase 1 in the cytoplasm, nucleus, cell membrane and cell junction. Tissue expression noted by UniProt (human): Appears to be expressed ubiquitously. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
