Product Overview

This rabbit polyclonal antibody recognizes pancreas/duodenum homeobox protein 1 (gene PDX1) in human samples and is validated for Western blot, flow cytometry and ELISA. The predicted molecular weight is 30.8 kDa, and the supplier reports an observed band at about 42 kDa.

PDX1 (also IPF1) is a homeodomain transcription factor essential for pancreas development. It specifies early pancreatic progenitors and, in adult β-cells, drives expression of insulin and other genes needed for glucose-regulated insulin secretion, so it is a key marker in β-cell biology, diabetes research and stem cell differentiation.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Samples with a confirmed signal (flow cytometry): 293T cells.

Reactivity Notes

The supplier lists reactivity with human. UniProt places pancreas/duodenum homeobox protein 1 in the nucleus and cytoplasm. Tissue expression noted by UniProt (human): Duodenum and pancreas (Langerhans islet beta cells and small subsets of endocrine non-beta-cells, at low levels in acinar cells). The samples tested by the supplier (listed above) are a practical starting point for positive controls.
