Key elements and design rationale

  • Target: CASP9 (Caspase-9).
  • Antibody format: Monoclonal; clone GAH-3; isotype Rabbit IgG.
  • Host: Rabbit.
  • Species reactivity: Human (confirm in your model system with appropriate controls).

This description is intended to help interpret the antibody design and the biological context of the target using the fields provided in the catalog record, alongside general experimental considerations.

Biological background

CASP9 (protein: P2X purinoceptor 1) is a commonly studied target in molecular and cellular biology. Functional context (as provided): Involved in the activation cascade of caspases responsible for apoptosis execution. Binding of caspase-9 to Apaf- 1 leads to activation of the protease which then cleaves and activates caspase-3. Promotes DNA damage-induced apoptosis in a ABL1/c-Abl-dependent manner. Proteolytically cleaves poly (ADP- ribose) polymerase (PARP). Reported cellular localization context: Cytoplasm, cytosol. Nucleus. Cytosolic under unstressed conditions, translocates into the nucleus upon induction by electrophilic agents. Tissue expression notes (as provided): Ubiquitous, with highest expression in the heart, moderate expression in liver, skeletal muscle, and pancreas. Low levels in all other tissues. Within the heart, specifically expressed in myocytes. .

Research relevance and current trends

  • Research context keywords from the source record include: Apoptosis,Apoptotic Markers,Cancer,Caspases,Cell Biology,Cell Death,Intracellular,Invasion/Microenvironment,Metabolism,Metabolism Processes,Pathways and Processes.
  • Current studies often focus on connecting target abundance/localization to pathway perturbations across models, tissues, and cell states.
  • Quantitative and multiplexed assays (e.g., imaging + immunoblot panels) are commonly used to compare phenotypes across conditions and time-courses.

Common research applications

  • Western blotting (WB): assess relative target abundance across samples, treatments, or time-points.
  • Immunohistochemistry (IHC): evaluate spatial distribution of target-positive staining in tissue architecture.
  • Flow cytometry: quantify target-positive populations and compare shifts in marker distributions.

Workflow ideas (metafield): Validate CASP9 antibody specificity using KO/KD control samples (WB/IF/IHC as appropriate), Detect CASP9 expression by Western blot in cell or tissue lysates, Detect CASP9 in FFPE tissue sections by immunohistochemistry, Quantify CASP9-positive cells by flow cytometry in single-cell suspensions

Notes for experimental interpretation

  • Consider isoforms and post-translational modifications (PTMs) that may shift apparent molecular weight or epitope accessibility.
  • Apparent molecular weight may vary by sample type and processing (observed MW: 46 kDa; calculated MW: 46281 MW).
  • Control concepts: include appropriate negative controls (e.g., isotype, KO/KD samples) and orthogonal validation when feasible.

Additional product details (from the source record)

  • Molecular weight (observed): 46 kDa
  • Cellular localization (provided): Cytoplasm, cytosol. Nucleus. Cytosolic under unstressed conditions, translocates into the nucleus upon induction by electrophilic agents.
  • Tissue details (provided): Ubiquitous, with highest expression in the heart, moderate expression in liver, skeletal muscle, and pancreas. Low levels in all other tissues. Within the heart, specifically expressed in myocytes. .

We are currently using anti-pro Caspase 9 Rabbit Monoclonal antibody M00080-1 for human tissue, and we are well pleased with the WB results. The species of reactivity given in the datasheet says human. Is it likely that the antibody can work on pig tissues as well?
The anti-pro Caspase 9 Rabbit Monoclonal antibody (M00080-1) has not been tested for cross reactivity specifically with pig tissues, though there is a good chance of cross reactivity. We have an innovator award program that if you test this antibody and show it works in pig you can get your next antibody for free. Please contact me if I can help you with anything.
We have observed staining in human adrenal tissue. Are there any suggestions? Is anti-pro Caspase 9 Rabbit Monoclonal antibody supposed to stain adrenal tissue positively?
From what I have seen in literature adrenal tissue does express CASP9. From what I have seen in Uniprot.org, CASP9 is expressed in adrenal tissue, t-cell, stomach cancer, kidney, eye lymph, cervix carcinoma, cervix carcinoma erythroleukemia, among other tissues. Regarding which tissues have CASP9 expression, here are a few articles citing expression in various tissues: Cervix carcinoma, Pubmed ID: 18669648, 20068231 Cervix carcinoma, and Erythroleukemia, Pubmed ID: 23186163 Eye, and Lymph, Pubmed ID: 15489334 Kidney, Pubmed ID: 14702039 Stomach cancer, Pubmed ID: 9890966 T-cell, Pubmed ID: 8900201
