Biological background

Biological context: 3'-exoribonuclease that has a preference for poly (A) tails of mRNAs, thereby efficiently degrading poly (A) tails. Exonucleolytic degradation of the poly (A) tail is often the first step in the decay of eukaryotic mRNAs and is also used to silence certain maternal mRNAs translationally during oocyte maturation and early embryonic development. Interacts with both the 3'-end poly (A) tail and the 5'-end cap structure during degradation, the interaction with the cap structure being required for an efficient degradation of poly (A) tails. Involved in nonsense-mediated mRNA decay, a critical process of selective degradation of mRNAs that contain premature stop codons. Also involved in degradation of inherently unstable mRNAs that contain AU-rich elements (AREs) in their 3'-UTR, possibly via its interaction with KHSRP. Probably mediates the removal of poly (A) tails of AREs mRNAs, which constitutes the first step of destabilization.

  • Background: Prostate-specific antigen (PSA), also known as gamma-seminoprotein or kallikrein-3 (KLK3), is a glycoprotein enzyme encoded in humans by the KLK3 gene. KLK3 is a member of the kallikrein-related peptidase family and is secreted by the epithelial cells of the prostate gland. This gene is mapped to 19q13.33. KLK3 is present in small quantities in the serum of men with healthy prostates, but is often elevated in the presence of prostate cancer or other prostate disorders. KLK3 is produced for the ejaculate where it liquifies the semen in the seminal coagulum and allows sperm to swim freely. It is also believed to be instrumental in dissolving the cervical mucous cap, allowing the entry of sperm. It is not a unique indicator of prostate cancer, but may also detect prostatitis or benign prostatic hyperplasia.
