Product Overview
Clone HBH-16 is a rabbit monoclonal antibody that recognizes phosphatidylinositol 3,4,5-trisphosphate 3-phosphatase and dual-specificity protein phosphatase PTEN in human, mouse and rat samples and is validated for Western blot, IHC and ICC/IF. The predicted molecular weight is 47.2 kDa, and the supplier reports an observed band at about 54 kDa.
PTEN is a lipid and protein phosphatase that removes the 3-phosphate from PIP3, directly opposing PI3K and dampening AKT signaling. By restraining this pathway it limits cell growth, survival and proliferation, and it is one of the most frequently inactivated tumor suppressors in human cancer.

Samples with a confirmed band (WB): human HeLa cells, human HepG2 cells, rat brain tissue, rat PC-12 cells, mouse brain tissue, mouse NIH/3T3 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:1000 overnight at 4 °C, HRP-conjugated secondary antibody at 1:1000, ECL detection.
Samples with confirmed staining (IHC): human colon tissue, rat lung tissue, human colon cancer tissue, human tongue cancer tissue, human liver tissue, mouse skin tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Samples with confirmed staining (ICC/IF): HeLa cells.
Immunogen
Synthetic peptide based on the sequence of human PTEN.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places phosphatidylinositol 3,4,5-trisphosphate 3-phosphatase and dual-specificity protein phosphatase PTEN in the cytoplasm, nucleus, cell projection and postsynaptic density. Tissue expression noted by UniProt (human): Expressed at a relatively high level in all adult tissues, including heart, brain, placenta, lung, liver, muscle, kidney and pancreas. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
