Anti-S1PR3 (EDG3) (extracellular) Antibody (#ASR-013) is a highly specific antibody directed against an extracellular epitope of mouse sphingosine 1-phosphate receptor 3. The antibody can be used in western blot and indirect flow cytometry applications. It has been designed to recognize S1PR3 from human, rat and mouse samples. Anti-S1PR3 (EDG3) (extracellular)-ATTO Fluor-488 Antibody (#ASR-013-AG) is directly labeled with an ATTO-488 fluorescent dye. ATTO dyes are characterized by strong absorption (high extinction coefficient), high fluorescence quantum yield, and high photo-stability. The ATTO-488 label is analogous to the well known dye fluorescein isothiocyanate (FITC) and can be used with filters typically used to detect FITC. Anti-S1PR3 (EDG3) (extracellular)-ATTO Fluor-488 Antibody is especially suited for experiments requiring simultaneous labeling of different markers.

Target S1PR3

Overview

Anti-S1PR3 (EDG3) (extracellular)-ATTO Fluor-488 Antibody is an antibody targeting Sphingosine 1-phosphate receptor 3, S1P3, Endothelial differentiation G-protein coupled receptor 3, Lysophospholipid receptor B3, LPB3 Polyclonal raised in Rabbit (ATTO-488. Maximum absorption 501 nm; maximum fluorescence 523 nm. The fluorescence is excited most efficiently in the 480 - 515 nm range. This label is analogous to the dye fluorescein isothiocyanate (FITC) and can be used with filters used to detect FITC.). This antibody is commonly used in FC to detect, localize, or compare expression of the target across samples.

Key elements and design rationale

  • Target: Sphingosine 1-phosphate receptor 3, S1P3, Endothelial differentiation G-protein coupled receptor 3, Lysophospholipid receptor B3, LPB3 (also reported as Sphingosine 1-phosphate receptor 3, S1P3, Endothelial differentiation G-protein coupled receptor 3, Lysophospholipid receptor B3, LPB3).
  • Immunogen/epitope region: Extracellular, N-terminus.
  • Homology note: Rat - identical; human - 9/12 amino acid residues identical (informative for cross-species interpretation).
  • Species reactivity (as provided): Human, Rat, Mouse.
  • Lot quality control (as provided): Western blot analysis (unlabeled antibody, #ASR-013), and direct flow cytometry (labeled antibody)..
  • Peptide confirmation: Confirmed by amino acid analysis and mass spectrometry.
  • Blocking peptide: Available for antigen preadsorption control where appropriate.
  • Conjugate/format: ATTO-488. Maximum absorption 501 nm; maximum fluorescence 523 nm. The fluorescence is excited most efficiently in the 480 - 515 nm range. This label is analogous to the dye fluorescein isothiocyanate (FITC) and can be used with filters used to detect FITC. (may affect detection channel and background).

These attributes help researchers interpret whether signal reflects the intended target in a given assay and sample context.

Biological background

Sphingosine 1-phosphate (S1P) is an active byproduct of sphingomyelin metabolism. All cells can synthesize this biomolecule but the majority of its synthesis comes from erythrocytes and endothelial cells. In cases of inflammation, mast cells and platelets are the main source of sphingosine 1-phosphate1.

Research relevance and current trends

  • Comparing target expression across perturbations, genotypes, or treatment conditions.
  • Interpreting localization shifts alongside pathway or phenotypic readouts.
  • Using orthogonal controls (KO/KD, peptide competition, isotype concepts) to support conclusions.

Common research applications

  • Flow cytometry (direct/indirect): quantify target-positive populations and shifts in expression across subsets.

Interpretation typically benefits from comparing matched sample sets (e.g., treated vs control, WT vs KO/KD) and using orthogonal readouts where feasible.

Notes for experimental interpretation

  • Isoforms and post-translational modifications can shift apparent molecular weight or epitope accessibility across samples.
  • Cross-species signal may depend on epitope conservation; consult the provided homology note when selecting models.
  • Permeabilization, fixation, and antigen retrieval can change accessibility of intracellular vs extracellular epitopes.
  • Conceptual control: antigen preadsorption (blocking peptide) can help assess signal dependence on the immunogen region.
  • Provided control suggestions: Negative control: RIC-001-AG.
  • Application notes: see product-specific dilution/usage notes and control concepts provided in the dataset.
